隔药饼灸对高脂血症合并动脉粥样硬化兔ADPN、MMP-3的影响

摘　　要：目的:观察隔药饼灸对高脂血症合并动脉粥样硬化兔脂联素(A D P N)、基质金属蛋白酶-3(MMP-3)的影响。方法:取新西兰兔32只,随机分为正常组、模型组、隔药饼灸组、阿托伐他汀组,每组8只。给予高胆固醇饲料喂养以制作高脂血症模型。各组予相应方法干预。HE染色比较各组兔腹主动脉管壁结构,检测血脂[低密度脂蛋白胆固醇(LDL-C)、甘油三酯(TG)、总胆固醇(TC)]、肿瘤坏死因子-α(TNF-α)、ADPN、MMP-3含量。结果:正常组主动脉壁结构各层清晰,模型组内膜增厚,泡沫细胞聚集,两治疗组内膜稍增厚,内壁较平整。与正常组比较,模型组兔血清LDL-C、TG、TC、TNF-α、MMP-3水平均显著升高(P<0.01),ADPN水平显著下降(P<0.05);与模型组比较,两治疗组血清LDL-C、TG、TC、TNF-α水平均显著下降(P<0.05,P<0.01),ADPN水平均显著上升(P<0.05),隔药饼灸组血清MMP-3水平显著下降(P<0.01)。血清LDL-C、TG、TC、TNF-α水平与MMP-3含量呈正相关,与ADPN含量呈负相关(P<0.01,P<0.05)。结论:隔药饼灸可通过调节MMP-3、ADPN水平,抑制炎症反应,对动脉粥样硬化产生保护作用,且MMP-3、ADPN水平与血脂相关指标有明显相关性,可作为评价高脂血症疗效的新型指标。Objective:To observe the effects of herbal cake-separated moxibustion on the expression of adiponectin(ADPN)and matrix metallopeptidase-3(MMP-3)in the serum of rabbits with hyperlipidaemia and atherosclerosis.Methods:A total of 32 New Zealand rabbits were randomly divided into four groups,namely normal group,model group,herbal cakeseparated moxibustion group and atorvastatin group,8 rabbits in each group.The hyperlipidemia model was prepared by feeding a high cholesterol diet.Each group was given corresponding intervention methods.Observation on the morphology and structure of rabbit aortic wall by HE staining and the levels of lipidemia(LDL-C,TG,TC),TNF-α,ADPN and MMP-3 were detected.Results:The wall structure of rabbits in normal group was clear and the inner wall was smooth,while the wall structure of rabbits in model group was obviously damaged with thickened inner membrane and foam cells.The intima of two treatment groups was slightly thickened and the inner wall was relatively flat.LDL-C,TG,TC,TNF-α,MMP-3 in the model group were higher than those in the normal group(P<0.01),and the level of ADPN decreased(P<0.05);Compared with the model group,LDL-C,TG,TC,TNF-αin two treatment groups all decreased(P<0.05,P<0.01),the serum ADPN levels improved(P<0.05),the serum MMP-3 in herbal cake-separated moxibustion group were significantly lower(P<0.01).LDL-C,TG,TC and TNF-αcontent in serum were positively correlated with the levels of MMP-3,and negatively correlated with the levels of ADPN(P<0.01,P<0.05).Conclusion:Herbal cake-separated moxibustion may protect atherosclerosis in rabbits by regulating the levels of MMP-3 and ADPN,inhibiting inflammation.Moreover,the levels of MMP-3 and ADPN were significantly correlated with lipid related indexes,which could be used as new indicators to evaluate the therapeutic efficacy of hyperlipidemia.
