摘 要:目的:观察寿胎丸对复发性自然流产(RSA)大鼠血浆中代谢产物的影响,探讨其安胎作用相关的生物标志物及代谢通路。方法:用羟基脲+米非司酮造RSA模型,将40只大鼠随机分为正常妊娠组、模型组、黄体酮组和寿胎丸组,每组10只。观察大鼠胚胎丢失率,利用液质联用结合代谢组学方法对各组间的代谢差异进行表征。结果:与模型组比较,寿胎丸组能显著降低RSA大鼠胚胎丢失率(P<0.05),并上调RSA大鼠血浆孕酮、牛熊去氧胆酸、溶血脑磷脂等,下调3-氧-4,6-胆碱烯酸、胆甾烯酮、白三烯B4等发挥安胎止血作用。筛选出14条相关代谢通路,并发现BCAT1、GAPDH等基因可能是寿胎丸发挥保胎作用的潜在靶点。结论:寿胎丸可调节RSA大鼠血浆内多种代谢物水平和多种代谢通路,提示该复方具有多靶点、多通路的作用特点。Objective:To observe the effect of Shoutai Pills on the metabolites in plasma of recurrent spontaneous abortion(RSA)rats,and explore the possible biomarkers and metabolic pathways of Shoutai Pills in protecting fetus.Methods:Forty rats were randomly divided into normal pregnancy group,model group,progesterone group and Shoutai Pills group,hydroxyurea and mifepristone was used to create the RSA rats model.The embryo loss rate of rats was observed and the metabolic differences among the groups were characterized by liquid chromatography-mass spectrometry(LC-MS)strategy.Results:Compared with the model group,Shoutai Pills group significantly reduced the rate of embryo loss,and up regulated progesterone,tauroursodeoxycholic acid and lyso phosphatidyl ethanolamine,down regulated 3-OXO-4,6-choladienoic acid,cholestenone and leukotriene B4 in the plasma of RSA rats and play a role in preventing fetal bleeding.A total of 14 metabolic pathways related to the mechanism of Shoutai Pills were found.The genes including BCAT1 and GAPDH,etc.might be the potential biological targets of Shoutai Pills.Conclusion:Shoutai Pills could regulate the levels of multiple metabolites and metabolic pathways in the plasma of RSA rats,and have the characteristics of multiple targets and multiple pathways.
