摘 要:目的:分析元胡生物碱成分在元胡止痛片质量控制中的作用。方法:通过整理元胡相关文献,了解元胡的化学成分和药理作用,制定出元胡止痛片质量控制方案。使用多种色谱方法对元胡药材中化学成分进行分离并研究,再针对所得成分采用核磁共振、质谱以及物理化学性质进行结构鉴定,通过一测多评法测定元胡止痛片中的生物碱类成分和含量。结果:元胡药材共成功分离5种单体化合物,分别为去氢紫堇碱、d-紫堇碱、海罂粟碱、四氢巴马汀、四氢小檗碱。通过一测多评法测定4批元胡止痛片中上述生物碱成分,显示不同厂家生产的元胡止痛片中总生物碱含量差异明显。结论:通过多种色谱方法测定可以较为准确地出元胡药材中的有效生物碱成分,针对元胡止痛片采用一测多评法可以有效检测出不同生物碱成分和剂量,可作为元胡止痛片的质量控制方法,为进一步完善元胡止痛片质量标准提供科学的参考依据。Objective:To analyze the application effect of Yuanhu alkaloid component in quality control of Yuanhu painkillers.Methods:The chemical composition and pharmacological action of Yuanhu painkillers were investigated by collating related literatures,and the quality control plan of Yuanhu painkillers was formulated.A variety of chromatographic methods were used to separate and study the chemical composition of Yuanhu medicinal herbs,and then nuclear magnetic resonance,mass spectrometry and physical and chemical properties were used to identify the structure of the obtained components.The alkaloids component and content in Yuanhu painkillers were determined by one test and multiple evaluation methods.Results:Five monomer compounds were successfully isolated from Yuanhu medicinal herbs,which were dehydrocorydaline,D-corydaline,papaverine,tetrahydropalmatine and tetrahydroberberine.The contents of the above alkaloids components in 4 batches of Yuanhu painkillers were determined by one test and multiple evaluation methods.The results showed that the contents of total alkaloids in Yuanhu painkillers produced by different manufacturers were obviously different.Conclusion:Through a variety of chromatographic methods,the effective composition of alkaloids in Yuanhu medicinal herbs can be accurately determined,the different alkaloid compositions and dosage in Yuanhu painkillers can be determined by one test and multiple evaluation methods,which can be used for the quality control of Yuanhu painkillers and provide scientific reference for further improving quality standard of Yuanhu painkillers.
