摘 要:为了确定组织细胞因子在奶牛产后能量负平衡(NEB)诱导的卵巢静止(IO)发生中的作用。本试验根据产后21 d血浆β-羟丁酸(BHBA)和葡萄糖(Glu)的水平,从100头奶牛中选取能量正平衡(PEB)组和NEB组各30头。随后,根据产后50~60 d是否发情及卵泡发育状况,选取NEB组中IO奶牛(NEB-IO)和PEB组中发情奶牛(PEB-E)各6头进行采样。利用生化和ELISA方法分别检测血浆中代谢指标和细胞因子的水平;利用实时荧光定量PCR(qRT-PCR)和ELISA方法检测组织中细胞因子的水平。结果显示:与PEB组奶牛相比,NEB组奶牛产后21 d血浆中白细胞介素6(IL-6)极显著升高(P<0.01),成纤维细胞生长因子21(FGF21)和血管生成素样蛋白8(ANGPTL8)水平显著降低(P<0.05),并持续到产后50 d;3个细胞因子与BHBA、Glu和游离脂肪酸(NEFA)之间显著相关(P<0.05);与PEB-E组奶牛相比,NEB-IO组奶牛IL-6 mRNA表达量和蛋白含量在肌肉和卵巢中显著降低(P<0.05);FGF21和ANGPTL8 mRNA表达量和蛋白含量在肝脏中显著降低(P<0.05),但在卵巢中均无明显变化(P>0.05)。结果表明,奶牛产后NEB可通过调节细胞因子的变化在奶牛IO发生中发挥作用。In order to ascertain the role of tissue cytokines in the occurrence of inactive ovaries(IO)induced by postpartum negative energy balance(NEB)in dairy cows,this study selected 60 out of 100 cows based on the plasma levels ofβ-hydroxybutyric acid(BHBA)and glucose(Glu)at 21 days postpartum,and divided into positive energy balance(PEB)group(n=30)and NEB group(n=30).Then,according to the estrus performance and follicular development status in 50 to 60 days postpartum,6 IO cows in NEB group(NEB-IO)and 6 estrus cows in PEB group(PEB-E)were sacrificed and sampled.The concentrations of metabolites and cytokines in plasma were assessed by biochemical tests and ELISA,respectively,while the concentrations of cytokines in tissues were measured by quantitative real-time PCR(qRT-PCR)and ELISA.The results showed that compared with PEB cows,NEB cows had extremely higher concentrations of plasma interleukin-6(IL-6)(P<0.01)and lower concentrations of plasma fibroblast growth factor 21(FGF21)and angiopoietin-like protein 8(ANGPTL8)(P<0.05)at 21 days postpartum;these levels persisted until 50 days postpartum.The three cytokines were significantly correlated with BHBA,Glu and non-esterified fatty acids(NEFA)(P<0.05).Furthermore,compared with the cows of PEB-E group,in the cows of NEB-IO group,the mRNA and protein expression levels of IL-6 in muscle and ovary were significantly decreased(P<0.05);the mRNA and protein expression levels of FGF21 and ANGPTL8 in liver were significantly decreased(P<0.05),but displayed no significant difference in ovary(P>0.05).The results indicate that postpartum NEB could play a role in the occurrence of IO in dairy cows by regulating the changes of cytokines.
