莫西菌素浇泼驱虫剂对靶动物绵羊的安全性评价

摘　　要：为了客观评价莫西菌素(MXD)浇泼剂对靶动物羊的临床应用安全性,本试验将40只体重为(25.0±2.7)kg的断奶羔羊随机分为4个组,分别为不给药对照组(CTR组)、1倍剂量浇泼给药组[0.5 mg/(kg·bw),1×组]、3倍剂量浇泼给药组[1.5 mg/(kg·bw),3×组]和5倍剂量浇泼给药组[2.5 mg/(kg·bw),5×组]。给药第1天(1 d)按前述分组方法和剂量对各组羊只沿背中线浇泼给药,连续给药3 d,试验观察期为7 d,观察各组羊只临床表现和体重变化;测定给药前1天(0 d)和给药后第7天(7 d)各组羊只体温、脉搏和呼吸(TPR)、血液生理生化和尿常规指标;测量7 d时各组羊只脏器系数并采取组织样品制备石蜡切片观察组织病理学变化,综合评价MXD浇泼剂临床用药安全性。结果显示:与CTR组相比,各浇泼给药组羊只临床表现均正常,体重和TPR指标均无显著差异(P>0.05);3×组的血糖(GLU)和5×组的红细胞容积(MCV)、肌酐(CREA)均存在极显著差异(P<0.01),但均在正常参考值内,其他指标均无显著差异(P>0.05);各浇泼给药组尿常规指标无显著差异(P>0.05),白细胞、蛋白质、隐血、酮体、胆红素、葡萄糖均为阴性;各浇泼给药组脏器系数无显著差异(P>0.05),器官组织均无病理变化。本试验结果表明,MXD浇泼剂的使用安全范围较大,以5倍临床推荐剂量[2.5 mg/(kg·bw)]连续使用3 d,对靶动物羊无明显的毒副作用。To evaluate the safety of pour-on agent of moxidectin(pour-on-MXD)in target animal sheep,this study randomly divided 40 sheep with average weight about(25.0±2.7)kg into 4 groups,namely the control group(CTR group),the dose group[0.5 mg/(kg·bw),1×group],the triple dose group[1.5 mg/(kg·bw),3×group],and the quintuple dose group[2.5 mg/(kg·bw),5×group].The pour-on-MXD was poured on along the midline of sheep back for first 3 days according the above grouping,the experiment lasting for 7 days,and clinical manifestations and body weight changes in sheep were observed.The body temperature,pulse and respiration(TPR),the blood physiological and biochemical parameters,and urine routine indexes were assessed at one day before experiment(0 d)and at day 7 of the experiment(7 d).Then,at 7 d,the organ coefficients of each group were measured,the histological samples were collected,and the slides were H&E stained to observe the histopathological changes and evaluate the safety of the pour-on-MXD on target animals.The results showed that in comparison with the CTR group,all animals in different pour-on-MXD treated groups were clinically normal,with no significant differences in body weight and TPR(P>0.05).GLU in 3×group and both mean corpuscular volume(MCV)and creatinine(CREA)in 5×group were significantly increased(P<0.01),but still fall within the normal reference values,others were no significantly different from CTR group(P>0.05).There were no significant difference in urine routine indexes(P>0.05),white blood cells,protein,occult blood,ketone body,bilirubin and glucose were all negative.There were no significant difference in organ coefficients(P>0.05)and no significant histopathologic changes were observed.Thus,the pour-on-MXD has high safety margin and causes no significant toxic and side effects on sheep;the optimal usage dose for sheep is suggested as 5 times of the clinically recommended dose[2.5 mg/(kg·bw)]for 3 consecutive days.
