摘 要:目的研究醒神解郁汤在卒中后抑郁(PSD)大鼠模型中,对脑组织中proBDNF/mBDNF平衡以及血清中IL-17表达的影响,探究其影响神经损伤可能的作用机制。方法65只SD大鼠随机分为假手术组、卒中后抑郁组、醒神解郁汤低/高剂量组以及舍曲林组。假手术组在大脑中动脉缺血再灌注(MCAO)术中只分离动脉不做阻断。卒中后抑郁组在MCAO术后施加慢性温和性不可预见性应激压力(CUMS)以建立PSD模型;PSD建模成功后,分别以醒神解郁汤低剂量(18.72 g·kg^(-1)·d^(-1))、高剂量(37.44 g·kg^(-1)·d^(-1))及舍曲林(5.142 g·kg^(-1)·d^(-1))灌胃,连续14 d。记录大鼠强迫游泳静止不动时间,利用透射电镜观察大鼠海马组织结构,分别采用Wester blotting、ELISA检测大鼠脑组织proBDNF/mBDNF水平及血清IL-17水平。结果醒神解郁汤可明显改善卒中后抑郁模型大鼠海马组织神经损伤。与假手术组相比,卒中后抑郁组强迫游泳静止不动时间、IL-17水平、proBDNF/mBDNF比值均显著增加,差异有统计学意义(P<0.05)。醒神解郁汤低、高剂量组与卒中后抑郁组相比,强迫游泳静止不动时间、IL-17水平、proBDNF/mBDNF比值均明显下降,差异有统计学意义(P<0.05)。结论醒神解郁汤可通过调控大鼠proBDNF/mBDNF比值及血清IL-17表达,改善卒中后抑郁大鼠的神经损伤。Objective To study the effects of Xingshen Jieyu decoction on proBDNF/mBDNF balance in brain tissue and the expression of serum IL-17 in post-stroke depression(PSD)model rats,and explore the possible mechanism of its influence on nerve injury.Methods 65 SD rats were divided randomly into four groups:sham-operation group,PSD group,low-doses group and high-doses group of Xingshen Jieyu decoction and sertraline group.In the sham-operation group,only the artery was isolated during middle cerebral artery ischemia-reperfusion(MCAO)without blocking.The rats in other groups were exposed to chronic unpredictable mild stress after MCAO operation to establish PSD model.After PSD modeling was successful,low dose of Xingshen Jieyu decoction(18.72 g·kg^(-1)·d^(-1)),high dose of Xingshen Jieyu decoction(37.44 g·kg^(-1)·d^(-1))or sertraline(5.142 g·kg^(-1)·d^(-1))were administered by gavage for consecutive 14 days,respectively.The immobile time of forced swimming test was recorded.The structure of hippocampus of rat was observed under transmission electron microscope.The levels of proBDNF/mBDNF in brain tissue and IL-17 in serum of rats were measured by Wester blotting and ELISA,respectively.Results Xingshen Jieyu decoction could alleviate nerve injury in hippocampus of post-stroke depression model rats.Compared with the sham-operation group,the immobile time of forced swimming test,IL-17 level and proBDNF/mBDNF ratio in the PSD group increased significantly,with statistical significance(P<0.05).Compared with the PSD group,the immobile time of forced swimming test,IL-17 level and proBDNF/mBDNF ratio in the low-dose and high-dose Xingshen Jieyu decoction group decreased significantly,with statistical significance(P<0.05).Conclusion Xingshen Jieyu decoction can alleviate the nerve injury in hippocampus by regulating the ratio of proBDNF/mBDNF and the expression of serum IL-17 in PSD rats.
