摘 要:目的:分析人参皂苷Rg3对低密度脂蛋白(LDL)诱导人肾小球系膜细胞(HMC)转化生长因子-β/Smad/核因子kappa B(TGF-β/Smad/NF-κB)通路及细胞凋亡的影响。方法:将HMC细胞分为对照组(不做任何处理)、LDL组(40μg/ml的LDL处理)、人参皂苷Rg3低浓度组(LDL 40μg/ml+人参皂苷Rg315μmol/L)、人参皂苷Rg3中浓度组(LDL 40μg/ml+人参皂苷Rg330μmol/L)、人参皂苷Rg3高浓度组(LDL 40μg/mL+人参皂苷Rg360μmol/L)。采用MTT法检测HMC细胞增殖活性;Western Blotting法检测通路相关蛋白TGF-β、Smad2、Smad3、NF-κB以及B淋巴细胞瘤基因相关x蛋白(Bax)、B淋巴细胞瘤基因-2(Bcl-2)和半胱天冬酶-3(Caspase-3)细胞凋亡相关蛋白的表达水平。结果:与对照组相比,LDL组HMC细胞增殖活性、TGF-β、Smad2、Smad3、NF-κB蛋白表达水平明显升高,差异均有统计学意义(P<0.01);与LDL组相比,人参皂苷Rg3低、中、高浓度组HMC细胞增殖活性、TGF-β、Smad2、Smad3、NF-κB、Bcl-2蛋白表达水平明显降低,HMC细胞凋亡率、Bax和Caspase-3的蛋白水平明显升高,并呈浓度依赖性,差异均有统计学意义(P<0.01)。结论:人参皂苷Rg3可抑制LDL诱导的HMC细胞增殖,促进其凋亡,其作用机制可能与TGF-β/Smad/NF-κB信号通路密切相关。Objective:To investigate the effects of ginsenoside Rg3on transforming growth factor-β/Smad/nuclear factor kappa B(TGF-β/Smad/NF-κB)pathway and apoptosis of human mesangial cells(HMC)induced by low density lipoprotein(LDL).Method:The cultured HMC cells were divided into the control group(without any treatment),the LDL group(40μg/ml LDL treatment),the low concentration group(LDL 40μg/ml+ginsenoside Rg315μmol/L),and the medium concentration group(LDL 40μg/ml+ginsenoside Rg330μmol/L group),high concentration group(LDL 40μg/ml+ginsenoside Rg360μmol/L).The proliferation activity of HMC cells was detected by MTT method,the expression levels of cell cycle related proteins of TGF-β,Smad2,Smad3,NF-κB,and apoptosis related proteins of B-lymphoma gene-related X protein(Bax),B-lymphoma gene-2(Bcl-2)and caspase-3were detected by Western Blotting.Results:Compared with those in the control group,the proliferation activity,TGF-β,Smad2,Smad3,NF-κB proteins expression levels of HMC cells in LDL group were significantly higher,and the differences were statistically significant.Compared with those in LDL group,the proliferation activity,TGF-β,Smad2,Smad3,NF-κB,Bcl-2proteins expression levels of HMC cells in low,medium and high concentration ginsenoside Rg3groups were significantly lower,the apoptosis rate of HMC cells and the protein levels of Bax and Caspase-3were significantly higher,with a concentration dependent manner,and the differences were statistically significant(P<0.01).Conclusion:Ginsenoside Rg3can inhibit the proliferation and promote the apoptosis of HMC cells induced by LDL,and its mechanism may be closely related to TGF-β/Smad/NF-κB signal pathway.
