摘  要:目的建立黔产香榧假种皮的高效液相色谱(HPLC)指纹图谱及多指标含量测定方法,为黔产香榧假种皮的资源利用提供科学依据。方法采用Agilent Eclipse Plus C 18色谱柱(4.6 mm×150 mm,5μm);以甲醇(A)-0.06%醋酸-水溶液(B)为流动相,梯度洗脱;流速1.0 mL/min;检测波长是230、255、270、330 nm;柱温30℃;进样量5μL。结果18批香榧假种皮指纹图谱相似度均大于0.995,共确定18个共有峰,指认了苯甲酸和金松双黄酮。对样品中的苯甲酸、异鼠李素、芹菜素、金松双黄酮和异银杏素5种化学成分进行含量测定和方法学考察,包括专属性、线性、精密度、重复性和稳定性,均符合要求;平均回收率在96.98%~101.08%,RSD≤1.57%。香榧假种皮中该5种化学成分的含量分别为(56.49~110.91、183.14~418.64、69.81~104.91、105.59~197.17、270.36~400.22)μg/g。结论本研究所建立的HPLC指纹图谱和多指标含量测定的方法准确可行,可作为黔产香榧假种皮质量评价的一种有效手段。Objective Establishment of fingerprinting and multi-component content determination methods by high performance liquid chromatography(HPLC)of the Torreya grandis aril,providing the scientific basis for resource utilization of Torreya grandis aril.Methods Agilent Eclipse Plus C 18 column(4.6 mm×150 mm,5μm)was used.With methanol(A)-0.06%acetic acid-water solution(B)as mobile phase,gradient elution was performed.The flow rate was 1.0 ml/min.The detection wavelengths were 230,255,270 and 330 nm.The column temperature was 30℃.The injection volume was 5μl.Results The similarity of 18 batches of Torreya grandis aril fingerprints were all greater than 0.995.A total of 18 common peaks were identified,and benzoic acid and sciadopitysin were identified.The methodological investigations were carried out on the determination of benzoic acid,isorhamnetin,apigenin,sciadopitysin,and isoginkgetin in the samples,including specificity,linearity,precision,repeatability,and stability,and all met the requirements.The average recoveries ranged from 96.98%to 101.08%,RSD≤1.57%.The contents of these five chemical components in Torreya grandis aril were(56.49-110.91,183.14-418.64,69.81-104.91,105.59-197.17,270.36-400.22)μg/g,respectively.Conclusion The method established by this research is accurate and feasible,and can be used as an effective means for quality evaluation of Torreya grandis aril.
