摘 要:目的基于核桃楸皮不同极性部位高效液相色谱(HPLC)指纹图谱与抗肝癌作用的谱效关系,筛选出核桃楸皮治疗肝癌的药效物质,揭示其治疗肝癌的药效物质基础。方法采用高效液相色谱法建立核桃楸皮不同极性部位的指纹图谱;采用CCK-8法开展核桃楸皮不同极性部位治疗肝癌的体外药效学研究;采用灰色关联度分析法及偏最小二乘回归分析法,将指纹图谱共有峰峰面积与药效学数据相关联,建立核桃楸皮不同极性部位治疗肝癌体外谱效关系,筛选出其治疗肝癌的药效物质,并进行体外药效验证。结果不同极性部位指纹图谱中共24个共有峰,其中有8个峰与药效呈正相关且变量投影(VIP)值大于1,确认这8种成分为核桃楸皮治疗肝癌的药效物质。经对照品比对,指认出4号峰为丁香酸、9号峰为鞣花酸、10号峰为槲皮苷、16号峰为山柰酚。结论本研究通过建立谱效关系筛选出了核桃楸皮治疗肝癌的药效物质,揭示了核桃楸皮治疗肝癌的药效物质基础。Objective Based on the spectrum-effect relationship between HPLC fingerprints of different polar parts of Cortex Juglans mandshurica and its anti-hepatocarcinoma effect,the pharmacodynamic substances of Cortex Juglans mandshurica for the treatment of hepatocarcinoma were screened out,and the pharmacodynamic material basis of Cortex Juglans mandshurica for the treatment of hepatocarcinoma was revealed.Methods HPLC method was used to establish the fingerprint of different polar parts of Cortex Juglans mandshurica,and CCK-8 method was used to study the pharmacodynamics of different polar parts of Cortex Juglans mandshurica on liver cancer in vitro in order to establish the in vitro spectral response relationship of different polar parts o Cortex Juglans mandshurica for the treatment of liver cancer,the common peak area of the fingerprint was correlated with the pharmacodynamic data by using gray correlation degree analysis and partial least squares regression analysis,the pharmacodynamic substances for the treatment of liver cancer were screened out and their pharmacodynamic effects were verified in vitro.Results There were 24 common peaks in the fingerprint of different polar parts,and 8 of them showed positive correlation with the drug effect and the VIP value was greater than 1.It was confirmed that these 8 components were the drug effective substances of Cortex Juglans mandshurica for treating liver cancer.The results showed that peak No.4 was syringic acid,peak No.9 was ellagic acid,peak No.10 was quercetin and peak No.16 was kaempferol.Conclusion In this study,the pharmacodynamic substances of Cortex Juglans mandshurica in the treatment of liver cancer were screened by establishing the spectrum-effect relationship,and the pharmacodynamic material basis of Cortex Juglans mandshurica in the treatment of liver cancer was revealed.
