摘 要:目的 研究银杏黄酮对川崎病小鼠冠状动脉损伤和核因子-κB(NF-κB)信号的影响。方法 实验分成对照组、模型组、实验低剂量组(100 mg·kg^(-1)银杏黄酮)、实验高剂量组(200 mg·kg^(-1)银杏黄酮)、阿司匹林组(45.5 mg·kg^(-1)阿司匹林)。取小鼠冠状动脉,用苏木精-伊红(HE)染色评价炎症细胞数目,用酶联免疫吸附(ELISA)法检测白细胞介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)、白细胞介素-1β(IL-1β)含量,用蛋白质印迹法检测自噬体吞噬蛋白(Beclin-1)、LC3-Ⅱ/LC3-Ⅰ、B细胞淋巴瘤-2(Bcl-2)、Bcl-2相关X蛋白(Bax)、p65蛋白表达。结果 对照组、模型组、实验低剂量组、实验高剂量组、阿司匹林组冠状动脉组织中炎症细胞数目分别为(1 054.62±148.49),(3 451.87±461.27),(2 647.51±322.48),(1 950.48±291.82),(2 028.33±195.46)cell·mm-2;IL-6含量分别为(60.84±5.17),(105.62±11.42),(92.10±7.13),(79.23±6.54),(72.91±7.84)pg·mL^(-1);TNF-α含量分别为(82.17±7.25),(99.12±6.33),(93.21±4.69),(86.05±8.25),(87.28±7.11)pg·mL^(-1);IL-1β含量分别为(79.24±6.33),(93.25±7.15),(85.47±8.14),(77.93±6.12),(80.56±7.84)pg·mL^(-1);Beclin-1蛋白表达水平分别为0.32±0.04,0.20±0.02,0.23±0.03,0.26±0.02,0.25±0.03;LC3-Ⅱ/LC3-Ⅰ蛋白表达水平分别为2.16±0.22,1.13±0.16,1.35±0.12,1.50±0.14,1.62±0.17;Bax蛋白表达水平分别为0.36±0.05,0.65±0.07,0.52±0.05,0.42±0.06,0.43±0.07;Bcl-2蛋白表达水平分别为0.66±0.06,0.40±0.05,0.45±0.03,0.49±0.04,0.51±0.06;p65蛋白表达水平分别为0.46±0.06,0.95±0.08,0.78±0.10,0.65±0.08,0.56±0.07,上述指标,对照组和模型组相比,差异均有统计学意义(均P<0.05);实验低剂量组、实验高剂量组、阿司匹林组与模型组相比,差异均有统计学意义(均P<0.05);实验高剂量组与实验低剂量组相比,差异均有统计学意义(均P<0.05)。结论 银杏黄酮改善川崎病小鼠冠状动脉炎症损伤,提高细胞自噬水平,减少细胞凋亡,机制可能与�Objective To investigate the effect of Ginkgo flavonoids on coronary artery injury and nuclear factor-κB (NF-κB) signal in mice with Kawasaki disease.Methods The experiment was divided into control group,model group,experimental low-dose group (100mg·kg^(-1)Ginkgo flavonoids),experimental high-dose group (200mg·kg^(-1)Ginkgo flavonoids),and aspirin group (45.5 mg·kg^(-1)aspirin).The coronary arteries of mice were taken;the number of inflammatory cells were detected by hematoxylineosin (HE) staining;the content of interleukin-6(IL-6),tumor necrosis factor-α(TNF-α),interleukin-1β(IL-1β) were detected by enzyme-linked immunosorbent assay (ELISA) method;the protein expression of autophagosome phagocytosis protein-1 (Beclin-1),LC3-Ⅱ/LC3-Ⅰ,B cell lymphoma-2 (Bcl-2),Bcl-2associated X protein(Bax),and p65 were detected by Western blot method.Results The number of inflammatory cells in the coronary artery tissues in control group,model group,experimental low-dose group,experimental highdose group and aspirin group were (1 054.62±148.49),(3 451.87±461.27),(2 647.51±322.48),(1 950.48±291.82),(2 028.33±195.46) cell·mm-2;the content of IL-6 were (60.84±5.17),(105.62±11.42),(92.10±7.13),(79.23±6.54),(72.91±7.84) pg·m L-1;TNF-α content were(82.17±7.25),(99.12±6.33),(93.21±4.69),(86.05±8.25),(87.28±7.11) pg·m L-1;IL-1β content were (79.24±6.33),(93.25±7.15),(85.47±8.14),(77.93±6.12),(80.56±7.84) pg·m L-1;Beclin-1protein expression levels were 0.32±0.04,0.20±0.02,0.23±0.03,0.26±0.02,0.25±0.03;LC3-Ⅱ/LC3-Ⅰprotein expression levels were 2.16±0.22,1.13±0.16,1.35±0.12,1.50±0.14,1.62±0.17;Bax protein expression levels were 0.36±0.05,0.65±0.07,0.52±0.05,0.42±0.06,0.43±0.07;Bcl-2 protein expression levels were 0.66±0.06,0.40±0.05,0.45±0.03,0.49±0.04,0.51±0.06;p65 protein expression levels were0.46±0.06,0.95±0.08,0.78±0.10,0.65±0.08,0.56±0.07,the above indicators,compared between control group and model group,the difference were all statistically significant (all P<0.05);
