摘  要:目的 探讨降糖三黄片改善糖尿病小鼠糖脂代谢紊乱的作用机制。方法 24只雄性db/db小鼠(自发糖尿病小鼠模型)随机分成模型组、降糖三黄片低、高剂量组(1.24g/kg、2.48g/kg)和阳性对照二甲双胍组(0.248g/kg),每组6只,同时将6只同窝db/m小鼠为正常组来作对照,待db/db小鼠血糖水平符合糖尿病模型后,药物干预各组按照上述剂量灌胃给药,每天给药1次,连续6周,正常组和模型组给予同等体积生理盐水灌胃。干预前后分别检测各组小鼠体重、摄食量、饮水量、空腹血糖及血脂代谢水平;使用Western Blot法检测小鼠胰腺组织自噬相关蛋白LC3B、P62及Beclin1的表达。结果 与正常组相比,模型组小鼠的体重、饮水量、摄食量、空腹血糖水平、TCHO、TG、LDL-C均明显升高(P<0.05),HDL-C显著降低(P<0.05)。与模型组相比,降糖低、高剂量组、二甲双胍组小鼠的体重、饮水量、摄食量、空腹血糖、TCHO、TG、LDL-C均显著下降(P<0.05),HDL-C显著上升(P<0.05)。蛋白表达方面,与正常组相比,模型组小鼠P62蛋白均明显升高(P<0.05),LC3B蛋白、Beclin1蛋白均显著降低(P<0.05);与模型组相比,降糖低、高剂量组小鼠的P62蛋白均显著下降(P<0.05),LC3B蛋白、Beclin1蛋白均显著上升(P<0.05)。结论 降糖三黄片可能是通过促进胰岛细胞自噬来改善糖尿病小鼠糖脂代谢紊乱情况。Objective To explore the mechanism of Jiangtang Sanhuang tablet in improving the disorder of glucose and lipid metabolism in diabetic mice. Methods 24 male db/db mice(spontaneous diabetic mice model) were randomly divided into model group, low-dose and high-dose Jiangtang Sanhuang tablet groups(1.24g/kg and 2.48g/kg) and positive control metformin group(0.248g/kg), with 6 mice in each group. At the same time, 6 db/m mice in the same nest were taken as normal control group. After the blood glucose level of db/db mice met the diabetic model, the drug intervention groups were given the above dose by gavage, once a day for 6 weeks, the normal group and model group were given the same volume of normal saline for 6 weeks. The body weight, food intake, water intake, fasting blood glucose and lipid metabolism levels of mice in each group were detected before and after the intervention;the expressions of autophagy related proteins lc3b, p62 and Beclin1 in islet cells were detected by Western blot. Results Compared with the normal group, the body weight, water intake, food intake, fasting blood glucose level, TCHO, TG, LDL-C of the model group were significantly increased(P<0.05), and HDL-C was significantly decreased(P<0.05). Compared with the model group, the body weight, water intake, food intake, fasting blood glucose, TCHO, TG and LDL-C of the mice in the hypoglycemic low-dose, high-dose and metformin groups were significantly decreased(P<0.05), while HDL-C was significantly increased(P<0.05). Compared with the normal group, p62 protein in the model group was significantly increased(P<0.05), lc3b protein and Beclin1 protein were significantly decreased(P<0.05);Compared with the model group, p62 protein in the low-dose and high-dose hypoglycemic groups were significantly decreased(P<0.05), lc3b protein and Beclin1 protein were significantly increased(P<0.05).Conclusion Jiangtang Sanhuang tablet may improve the disorder of glucose and lipid metabolism in diabetic mice by promoting the autophagy of islet cells.
