摘 要:目的:对杜鹃兰叶总黄酮的提取工艺进行优化,并研究其抗氧化活性。方法:采用超声辅助提取杜鹃兰叶总黄酮,考察单因素水平,结合响应面分析法对杜鹃兰叶总黄酮提取工艺进行优化。在采用三个单因素水平基础上,以乙醇浓度、液料比、提取时间为自变量,杜鹃兰叶总黄酮提取量为响应值,采用三因素三水平的Box-Behnken响应面法对杜鹃兰叶总黄酮的提取工艺进行优化。结果:当乙醇浓度为74%时,超声29 min,液料比为47∶1 mL/g,杜鹃兰叶总黄酮平均提取率可达到(24.34±1.37)mg/g,与预测值24.53 mg/g接近,偏差0.74%,同时,测得杜鹃兰叶提取液对DPPH和ABTS自由基的IC50分别为0.196、0.080 mg/mL。结论:优化后的提取工艺效果较好,杜鹃兰叶总黄酮具有较强的抗氧化作用。Objective To optimize the extraction technology of total flavonoids from cremastra appendiculata leaves and to evaluate their antioxidant activities.Methods Ultrasonic-assisted extraction of total flavonoids from cremastra appendiculata leaves was used to optimize the extraction process of total flavonoids from cremastra appendiculata leaves by combining response surface analysis with single factor level.On the basis of three single factor levels,taking ethanol concentration,liquid-to-material ratio and extraction time as independent variables,and the extraction amount of total flavonoids from rhododendron leaves as the response value.The Box-Behnken response surface method with three factors and three levels was used to optimize the extraction process of total flavonoids from rhododendron leaves.Results When the ethanol concentration was 74%,ultrasonication was performed for 29 minutes and the ratio of liquid to material was 47∶1 mL/g,the average extraction rate of total flavonoids from Rhododendron leaves could reach(24.34±1.37)mg/g,which was very close to the predicted value of 24.53 mg/g,with a deviation of 0.74%.At the same time,the measured IC50of cremastra appendiculata leaves extract on DPPH and ABTS free radicals were 0.196 and 0.080 mg/mL,respectively.Conclusion The optimized extraction process is better,and the total flavonoids of cremastra appendiculata leaves have strong antioxidant effect.
