摘  要:目的:探讨lncRNA-NR_046269对马尔尼菲篮状菌(TM)感染人支气管上皮细胞早期IL-6表达的影响。方法:实验组和对照组分别用TM孢子和RPMI1640培养基与BEAS-2B细胞株共孵育4 h,生物信息学方法筛选NR_046269/IL-6关系对子,采用慢病毒载体转染BEAS-2B细胞株,分别敲低和过表达NR_046269,验证NR_046269对IL-6有无调控作用。结果:生物信息学分析显示,NR_046269与IL-6呈显著负相关(PCC=-0.998,P<0.05)。qPCR和ELISA结果均显示实验组IL-6表达显著降低(P<0.05)。NR_046269敲低组IL-6表达显著升高,而NR_046269过表达组IL-6表达显著降低(P<0.05)。结论:人支气管上皮细胞在TM感染早期IL-6表达显著降低。通过负调控IL-6表达,lncRNA-NR_046269有望成为临床治疗TM的新靶点。Objective:To investigate effect of lncRNA-NR_046269 on expression of IL-6 in human bronchial epithelial cells response to early stage infection of Talaromyces marneffei(TM).Methods:Treated and control groups were co-cultured with TM spores and RPMI1640 medium and BEAS-2B cells for 4 h,respectively.Correlationship pair of NR_046269/IL-6 was screened by bioinformatics.BEAS-2B cells were transfected with lentiviral vector to interference or overexpression of NR_046269,so as to confirm whether NR_046269 had an regulatory effect on IL-6.Results:Bioinformatics analysis results showed that NR_046269 was negatively correlated with IL-6(PCC=-0.998,P<0.05).Results of qPCR and ELISA showed that expression of IL-6 were decreased in treated group(P<0.05).Knockdown of NR_046269 enhanced IL-6 mRNA expression,whereas overexpression of NR_046269 decreases IL-6mRNA expression(P<0.05).Conclusion:In early stage of TM infection,expression of IL-6 in human bronchial epithelial cells is down-regulated.Through down-regulating IL-6 expression,lncRNA-NR_046269 is expected to become a new target for clinical treatment of TM.
