摘  要:目的:探究T辅助细胞17(Th17)/调节性T细胞(Treg)介导炎症反应对糖尿病(DM)大鼠动脉内皮细胞凋亡影响机制。方法:腹腔注射链脲佐菌素建立大鼠DM模型,将DM大鼠分为DM组和治疗组(n=12),另取12只健康大鼠作为对照组。比较各组大鼠的血糖、主动脉损伤情况、血清IL-6、TNF-α和动脉内皮细胞中凋亡标志蛋白Caspase3和Bcl-2水平。并通过流式细胞术检测外周血中Th17/Treg水平。结果:建模后大鼠血糖均高于16.7 mmol/L,干预后治疗组的空腹血糖水平显著低于DM组(P<0.05)。DM组的主动脉血管壁厚度(54.47±2.61)μm、血清IL-6(19.24±2.21)pg/mg和TNF-α(39.46±3.56)pg/mg水平、动脉内皮细胞中Caspase3 mRNA和Bcl-2 mRNA和蛋白表达量、Th17水平(2.72±0.38)%和Th17/Treg比例(1.23±0.19)显著高于对照组,Treg水平(2.21±0.38)%显著低于对照组(P<0.05)。治疗组动脉壁厚度(46.82±2.72)μm、IL-6(13.86±1.87 pg/mg、TNF-α(22.35±3.04)pg/mg、Caspase3 mRNA和Bcl-2 mRNA和蛋白表达量、Th17水平(1.50±0.20)%和Th17/Treg比例(0.46±0.07)显著低于DM组,Treg(3.26±0.52)%水平显著高于DM组(P<0.05)。结论:Th17/Treg免疫细胞介导炎症反应可能参与了DM引起的动脉内皮细胞凋亡。Objective:To investigate the mechanism of T help 17 cells(Th17)/regulatory T cells(Treg)mediated inflammatory response on the apoptosis of arterial endothelial cells in diabetes mellitus(DM)rats.Methods:The rat model of DM was established by intraperitoneal injection of streptozotocin.DM rats were divided into DM group and treatment group(n=12).Another 12healthy rats were taken as control group.The blood glucose,aortic injury,serum IL-6,TNF-αand the levels of apoptosis marker proteins Caspase3 and Bcl-2 in arterial endothelial cells were detected in each group of rats.The Th17/Treg levels in peripheral blood were detected by flow cytometry.Results:After modeling,the blood glucose of rats was above 16.7 mmol/L.After the intervention,the fasting blood glucose level of the treatment group was significantly lower than that of DM group(P<0.05).Aortic vessel wall thickness(54.47±2.61)μm,serum IL-6(19.24±2.21)pg/mg and TNF-α(39.46±3.56)pg/mg levels,Caspase3 mRNA and Bcl-2 mRNA and protein in arterial endothelial cells,Th17 level(2.72±0.38)%and Th17/Treg ratio(1.23±0.19)in DM group were significantly higher than those in control group,and the Treg level(2.21±0.38)%was significantly lower than that in control group(P<0.05).The arterial wall thickness of treatment group was(46.82±2.72)μm,IL-6(13.86±1.87)pg/mg,TNF-α(22.35±3.04)pg/mg,Caspase3mRNA and Bcl-2 mRNA and protein expression,Th17 level(1.50±0.20)%and Th17/Treg ratio(0.46±0.07)were significantly lower than those in DM group,and the Treg(3.26±0.52)%level was significantly higher than that in DM group(P<0.05).Conclusion:Th17/Treg immune cell-mediated inflammation may be involved in the apoptosis of arterial endothelial cells caused by DM.
