摘　　要：目的建立同时测定川西獐牙菜中马钱苷酸、獐牙菜苦苷、龙胆苦苷、当药苷、芒果苷、异荭草素、牡荆素、当药醇苷、1,8-二羟基-3,7-二甲氧基酮、8-O-β-D-吡喃葡萄糖-1,3,5-三羟基酮含量的高效液相色谱法。方法用Waters Sunfire ODS C 18柱(250 mm×4.6 mm,5μm),以甲醇-水(含0.5 mL·L^(-1)磷酸)为流动相,进行梯度洗脱,流速为1.0 mL·min^(-1),检测波长为254 nm,柱温为30℃。结果马钱苷酸、獐牙菜苦苷、龙胆苦苷、当药苷、芒果苷、异荭草素、牡荆素、当药醇苷、1,8-二羟基-3,7-二甲氧基酮、8-O-β-D-吡喃葡萄糖-1,3,5-三羟基酮均得到良好分离,线性关系良好(r≥0.9996)。加样回收率均在92.26%~100.78%,RSD≤1.75%。结论该方法具有良好的重复性和稳定性,对全面评价川西獐牙菜药材质量和充分开发利用药用资源提供了一定的参考依据。Objective To establish a method for simultaneous determination of loganic acid, swertiamarin, gentiopicroside, sweroside, mangiferin, isoorientin, vitexin, swertianolin, 1,8-dihydroxy-3,7-dimethoxyxanketone, and 8-O-β-D-glucopyranose-1,3,5-trihydroxyxanketone in Swertia mussotii Franch. by HPLC.Methods Waters Sunfire ODS C18column(250 mm ×4.6 mm, 5 μm) was used with methanol-water(containing 0.5 mL·L^(-1)phosphoric acid) as mobile phase in gradient elution mode. The flow rate was 1.0 mL·min^(-1), the detection wavelength was 254 nm and the column temperature was 30 ℃.Results Loganic acid, swertiamarin, gentiopicroside, sweroside, mangiferin, isoorientin, vitexin, swertianolin, 1,8-dihydroxy-3,7-dimethoxyxanketone, and 8-O-β-D-glucopyranose-1,3,5-trihydroxyxanketone were well separated and showed good linear relationships(r≥0.999 6). The recoveries were 92.26%-100.78%, RSD≤1.75%.Conclusion This method has good repeatability and stability. This study provides a reference for the comprehensive evaluation of the quality of Swertia mussotii Franch. and the full development and utilization of medicinal resources.
