基于网络药理学的养精种玉汤改善种植窗期子宫内膜容受性机制分析

摘　　要：目的 运用网络药理学方法分析养精种玉汤改善种植窗期子宫内膜容受性的分子机制。方法:借助TCMSP、Genecards等数据库,检索方中药物的有效成分、蛋白靶点及与子宫内膜容受状态密切相关的靶点基因;借助Venny R软件获取“药物-疾病”交互靶点;Cytoscape 3.7.2软件构建一体可视化网络图;基于David数据库的GO、KEGG分析获得药物成分发挥作用的关键生物学过程和信号通路。结果:该方共筛选出31种活性成分,其改善种植窗期子宫内膜容受性的交集靶点249个,核心靶点围绕STAT3、APP、IL6等调控的细胞增殖、炎症-免疫反应等生物学过程,主要涉及癌症、PI3K-AKT、FoxO、VEGF等相关通路。结论:养精种玉汤可通过改善内膜生长、分泌状态和局部血流灌注、平衡母胎界面免疫-炎症微环境,修复内膜损伤等多途径促进胚胎着床。Objective: The paper aims at analyzing the molecular mechanism of Yangjing Zhongyu Decoction in improving endometrial receptivity during implantation window period via network pharmacology. Methods: With the TCMSP database and Genecards databases, the active constituents and protein targets of Yangjing Zhongyu Decoction, as well as target genes related to endometrial receptivity were retrieved. The “drug-disease” interactive targets were obtained by Venny R software. With the Cytoscape 3.7.2 Software, integrated Visual Network Diagram was built. Biological processes and key signaling pathways related to the improvement of endometrial receptivity were acquired via GO analysis and KEGG analysis. Results: A total of 31 active ingredients of Yangjing Zhongyu Decoction were screened and 249 intersection targets were identified for improving endometrial receptivity during implantation window. The core targets were around biological processes such as cell proliferation, endometrial inflammation and immune response regulated by STAT3, APP and IL6. It is mainly involved in cancer, PI3K-AKT, FoxO, VEGF and other pathways. Conclusions: Yangjing Zhongyu Decoction promotes embryo implantation by improving endometrium growth, secretory and local blood supply, balancing immune-inflammatory microenvironment in maternal-fetal interface and repairing endometrium damage, etc.
