摘　　要：目的:建立HPLC法测定蕲蛇药酒中马钱苷酸和龙胆苦苷的含量。方法:采用Ultimate Plus C18色谱柱(250 mm×4.6 mm,5μm),流动相为乙腈(A)-0.1%磷酸溶液(B),梯度洗脱,流速为1.0 mL/min,检测波长为254 nm,柱温为30℃,进样量为10μL。结果:马钱苷酸和龙胆苦苷分别在浓度为0.01260 mg/mL~0.3149 mg/mL和0.02017 mg/mL~0.5042 mg/mL范围内呈良好的线性关系,平均加样回收率分别为100.19%、96.62%,RSD均小于2.0%。结论:HPLC法简便、准确,重复性好,可用于测定蕲蛇药酒中马钱苷酸和龙胆苦苷的含量。Objective:To establish a method for determining the content of loganic acid and gentiopicroside in agkistrodon medicinal liquor by HPLC.Methods:The determination was performed on Ultimate Plus C18 column(250 mm×4.6 mm,5μm)with mobile phase consisting of acetonitrile(A)-0.1%phosphoric acid(B)at the flow rate of 1.0 mL/min.The detection wavelength was 254 nm.The column temperature was 30℃.The injection volume was 10μL.Results:The results showed that loganin acid and gentiopicroside has a good linearity within 0.01260 mg/mL-0.3149 mg/mL and 0.02017 mg/mL-0.5042mg/mL,respectively.The average recoveries of loganic acid and gentiopicroside were 100.19%and 96.62%,respectively.The RSD was less than 2.0%.Conclusion:The methods is simple,accurate with a good reproducibility.It can effectively control the content of loganic acid and gentiopicroside in agkistrodon medicinal liquor.
