摘 要:目的通过测定天仙果根提取物各极性部位总黄酮含量,并评价其体外抗氧化抗炎活性与总黄酮含量的关系。方法用浓度为60%的乙醇对天仙果根黄酮类成分进行回流提取,分别用石油醚、乙酸乙酯、正丁醇、水萃取,得到4个不同极性组分;采用NaNO_(2)-Al(NO_(3))_(3)-NaOH法测定各组分总黄酮含量;采用DPPH、ABTS自由基清除及Fe^(3+)还原能力试验测定各组分抗氧化活性;构建LPS诱导RAW264.7释放NO的细胞模型及稳定转染核转录因子-κB(nuclear factor-κB,NF-κB)荧光素酶报告基因的人胚肾细胞(HEK293)模型评价各组分的抗炎活性。结果天仙果根提取物总黄酮含量(241.73±3.02)mg·g^(-1),各极性部位中,乙酸乙酯部位的总黄酮含量最高,为(764.00±7.37)mg·g^(-1);抗氧化、抗炎活性结果表明,天仙果提取物及各组分都具有一定的抗氧化抗炎活性,乙酸乙酯部位的活性最高,对DPPH、ABTS自由基的清除能力和Fe^(3+)还原能力的IC_(50)值分别为5.24、0.92和48.16μg·mL^(-1),对LPS诱导的RAW264.7细胞释放NO及TNF-α诱导的HEK293细胞NF-κB活化的抑制活性的IC_(50)值为48.68和33.19μg·mL^(-1)。结论天仙果不同组分间抗氧化抗炎活性的差异较大,其作用机制可能与多靶点、多成分、多途径相关,值得进一步研究。OBJECTIVE To determine the total flavonoids contents and evaluate the antioxidant and anti-inflammatory activities of different fractions of Ficus beecheyan extraction.METHODS The flavones were segregated from the root of Ficus beecheyan with 60%ethanol,and then extracted successively with petroleum ether,EtOAc,n-BuOH and water to get four fractions.Contents of total flavonoids in various fractions were determined by NaNO_(2)-Al(NO_(3))_(3)-NaOH assay.Antioxidant activity of the different fractions were evaluated by DPPH,ABTS free radical scavenging and Fe^(3+)reducing power measurement.And anti-inflammatory activity were evaluated by measuring NO that released from mice’s macrophage RAW264.7 induced by LPS and detecting the expression of TNF-αinduced activation of NF-κB in HEK293 cells,which were stably transfected with NF-κB luciferase reporter gene.RESULTS The content of total flavonoids of ethanol extraction of Ficus beecheyan roots was(241.73±3.02)mg·g^(-1).The content of total flavonoids of ethyl acetate fraction was(764.00±7.37)mg·g^(-1),which was significantly higher than that of any other fractions(P<0.001).The results revealed that the ethanol extraction of Ficus beecheyan and the different fractions had antioxidant and anti-inflammatory effect of different degree.Among them,the EtOAc fraction had the most potent anti-oxidative and anti-inflammatory effect.The IC_(50)values for EtOAc fraction against DPPH,ABTS free radical,and Fe^(3+)reducing power were 5.24、0.92 and 48.16μg·mL^(-1),respectively.The IC_(50)values of the inhibitory activities of LPS-induced NO release in RAW264.7 cells and TNF-αinduced NF-κB activation in HEK293 cells were 48.68 and 33.19μg·mL^(-1),respectively.CONCLUSION The antioxidant and anti-inflammatory activities show significant differences among different fractions of Ficus beecheyan.The mechanisms of Ficus beecheyan are related to multi-drugs,multi-targets and multi-pathways.It is required further in depth study.
