摘  要:目的探讨铜绿假单胞菌(PA)感染小鼠中耳后对内耳及毛细胞的损伤。方法选取出生后6~8周的健康CBA小鼠,分为对照组(PBS种植组)和PA左侧中耳种植组,每组小鼠再分为PBS/PA种植后饲养3天组、7天组、14天组;使用耳内镜、小动物体内成像系统、免疫荧光等方法对小鼠中耳、内耳感染情况及耳蜗毛细胞的损伤情况进行观察分析。结果PA感染中耳后3天、7天、14天中耳腔出现不同程度的黏膜充血水肿,鼓膜穿孔及积脓等化脓性中耳炎的症状;圆窗可见颗粒样物质沉积;小动物体内成像检查可检测到部分耳蜗出现PA感染信号;免疫荧光染色及细胞计数发现,PA感染后7天底转毛细胞计数88.33±66.40,感染后14天小鼠底转、中转毛细胞计数分别为85±33.45,166±33.41,均出现不同程度的缺失,与PBS对照组比较差异显著(P值均小于0.05)。结论PA感染小鼠中耳后细菌及其产物可进入内耳并引起耳蜗毛细胞的损伤;引起毛细胞损伤的可能是PA的慢性毒性。Objective To investigate damage of inner ears and hair cells following middle ear infection with Pseudomonas aeruginosa(PA)in mice.Methods Healthy CBA mice(6 to 8 weeks old)were selected to receive PBS(control group)or PA(study group)injection into the left middle ear.At 3,7 and 14 days after PBS/PA injection,otoendoscopy and the Small Animal in Vivo Imaging System were used to detect middle ear.Immunofluorescence staining was used to detect damage of the cochleae and hair cells.Results Mucosal congestion and edema,tympanic membrane perforation and purulent exudate in the middle ear cavity were seen at 3,7 and 14 days after PA injection.Particulate substance deposition was visible on the round window membrane.Cochlear infection can be detected by the Small Animals In Vivo Imaging System in some animals.Immunofluorescence staining of cochlea hair cells showed various degrees of hair cell loss in PA treated animals as compared to the PBS control group(P<0.05):88.33±66.40 in basal turn at 7d;85±33.45 and 166±33.41 in the basal and middle turn,respectively,at 14d.Conclusion In mice,bacteria and their products in middle ear PA infection can enter the inner ear and cause inner ear hair cells damage.Persisting toxicity from PA may be the cause of hair cell damage.
