摘 要:目的探讨有氧运动对二氧化硅纳米颗粒(SNP)染毒大鼠血液毒性的改善作用。方法将40只Wistar大鼠随机分为空白对照组、染毒组、低强度运动组、中强度运动组和高强度运动组,每组8只。除空白对照组外,其余组均采用非暴露式气管滴注法构建SNP染毒模型,空白对照组采用同等体积的生理盐水进行气管滴注。建模成功后,低强度运动组、中强度运动组大鼠分别在每次气管滴注的第2天游泳45 min、游泳90 min,高强度运动组大鼠分别在每次气管滴注的第2天上午和下午各游泳75 min,空白对照组、染毒组不进行运动干预。于首次气管滴注前及处死大鼠前称量大鼠体重,计算体重增加值。于最后一次运动24 h后采血,检测各组大鼠的血常规指标、血生化指标、超氧化物歧化酶(SOD)及羟脯氨酸水平。结果染毒组大鼠的白细胞计数、血小板计数升高,红细胞计数降低,血清ALT、AST、肌酐、尿素氮、乳酸脱氢酶、SOD水平均升高(均P<0.05)。低强度运动组、中强度运动组大鼠的体重增加值均低于空白对照组、染毒组、高强度运动组(均P<0.05)。与染毒组、低强度运动组相比,中、高强度运动组大鼠的白细胞计数和血小板计数降低,红细胞计数升高,且血清ALT、AST、肌酐、乳酸脱氢酶、SOD水平均下降(均P<0.05)。中强度运动组大鼠的血清羟脯氨酸水平低于染毒组、低强度运动组、高强度运动组(均P<0.05)。结论中、高强度的有氧运动可以缓解SNP造成的炎症反应和氧化应激反应,减轻SNP造成的心脏、肝脏及肾脏损害。Objective To explore the improvement of aerobic exercise on hematotoxicity of rats with silica nanoparticle(SNP)poisoning.Methods A total of 40 Wistar rats were randomly divided into blank control group,poisoning group,low-intensity exercise group,medium-intensity exercise group,or high-intensity exercise group,with 8 rats in each group.Except for the blank control group,SNP poisoning models were constructed in the remaining groups by the method of intratracheal instillation in non-exposed type,while the blank control group received intratracheal instillation of normal saline with equivalent volume.After successful modeling,rats of the low-and medium-intensity exercise groups swam for 45 and 90 minutes on the second day of each intratracheal instillation,respectively,rats of the high-intensity exercise group swam for 75 minutes in the morning and afternoon on the second day of each intratracheal instillation,and the blank control group and the poisoning group did not receive exercise intervention.The body weight of the rats was measured before the first intratracheal instillation and before killing rats,and the weight gain was calculated.Blood was collected 24 hours after the last exercise,and the blood routine indexes,blood biochemical indexes,superoxide dismutase(SOD)level and hydroxyproline level of rats were detected in various groups.Results The poisoning group exhibited elevated white blood cell counts and blood platelet counts,whereas decreased erythrocyte counts,and elevated levels of serum ALT,AST,creatinine,urea nitrogen,lactate dehydrogenase,and SOD(all P<0.05).The weight gains of rats in the low-and medium-intensity exercise groups were lower than those of rats in the blank control group,the poisoning group,and the high-intensity exercise group(all P<0.05).Compared with the poisoning and low-intensity exercise group,rats in the medium-and high-intensity exercise groups obtained decreased white blood cell counts and blood platelet counts,whereas elevated erythrocyte counts,and decreased levels of serum
