摘  要:目的:研究黄连素(BBR)对波动性高糖导致的人冠状动脉内皮细胞(HCAECs)炎症反应的作用。方法:分离、培养HCAECs,将HCAECs分为5组。正常血糖组(NG组)加入5.5 mmol/L葡萄糖;持续性高糖组(PHG组)加入25 mmol/L葡萄糖;波动性高糖组(IHG组)加入5.5 mmol/L葡萄糖和25 mmol/L葡萄糖24 h波动1次;对照组(OC组)加入5.5 mmol/L和25 mmol/L甘露醇24 h波动1次,为波动性高渗环境;波动性高糖+黄连素组(IHG+BBR组):5.5 mmol/L葡萄糖和25 mmol/L葡萄糖+50μmol/L黄连素24 h波动1次。以上5组HCAECs培养7 d。用酶联免疫吸附实验(ELISA)测定白细胞介素-1β(IL-1β)、肿瘤坏死因子-α(TNF-α)、人单核细胞趋化蛋白-1(MCP-1)和白细胞介素-10(IL-10)等炎症指标的蛋白水平,实时荧光定量聚合酶链式反应(qRT-PCR)测定IL-1β、TNF-α、MCP-1和IL-10的mRNA表达。结果:PHG组和IHG组IL-1β、TNF-α、MCP-1水平较NG组和OC组明显升高,IL-10表达水平较NG组和OC组明显降低(P<0.05)。IHG组IL-1β、TNF-α、MCP-1较PHG组升高明显,IL-10较PHG组降低明显(P<0.05)。IHG+BBR组IL-1β、TNF-α、MCP-1表达水平较IHG组降低,IL-10表达水平较IHG组升高(P<0.05)。结论:波动性高糖比持续性高糖更易引起HCAECs的炎症反应,黄连素对波动性高糖导致的HCAECs的炎症反应具有明显的保护作用。Objective:To investigate the effect of Berberine(BBR)on the inflammatory response of human coronary artery endothelial cells(HCAECs)induced by fluctuating high glucose.Methods:HCAECs were isolated and cultured,and then divided into 5 groups.Blood glucose group(NG group)was added with 5.5 mmol/L glucose.High glucose group(PHG group)was treated with 25 mmol/L glucose,which was continuous high glucose.Fluctuating high glucose group(IHG group)was treated with 5.5 mmol/L glucose and 25 mmol/L glucose successively once 24 hours.Control group(OC group)was treated with 5.5 mmol/L and 25 mmol/L mannitol successively once every 24 hours in a fluctuating hypertonic environment.High fluctuation sugar+Berberine group(IHG+BBR group)was treated with 5.5 mmol/L glucose and 25 mmol/L glucose+50μmol/L Berberine was fluctuated once 24 hours.HCAECs in the above 5 groups were cultured for 7 days.The protein levels of interleukin-1β(IL-1β),tumor necrosis factor-α(TNF-α),human monocyte chemokine protein-1(MCP-1),and interleukin-10(IL-10)were determined by enzyme-linked immunoadsorption assay(ELISA).The mRNA expressions of IL-1β,TNF-α,MCP-1,and IL-10 were determined by real-time fluorescence quantitative PCR(qRT-PCR).Results:Compared with NG and OC groups,the levels of IL-1β,TNF-α,and MCP-1 in PHG and IHG groups were significantly increased,and the expression level of IL-10 was significantly decreased(P<0.05).Compared with PHG group,the increase of IL-1β,TNF-α,and MCP-1 was more significant in IHG group,and the decrease of IL-10 was more significant(P<0.05).When Berberine was added to IHG group,the expression levels of IL-1β,TNF-α,and MCP-1 in IHG+BBR group were decreased compared with that in IHG group,and the expression level of IL-10 was increased(P<0.05).Conclusion:Fluctuating high glucose was more likely to cause the inflammatory response of HCAECs compared with persistent high glucose.Berberine showed an obvious protective effect on the inflammatory response of HCAECs caused by fluctuating high glucose.
