摘 要:为研究甜樱桃砧木WRKY71转录因子的功能及其对非生物胁迫和激素处理的响应,本研究克隆了甜樱桃砧木‘Gisela 6’的PcWRKY71基因序列,序列分析表明PcWRKY71完整开放阅读框为1 014 bp,编码337个氨基酸,含有1个WRKY保守结构域和1个C_(2)H_(2)锌指结构域,属于WRKY基因家族第Ⅱ类成员。生物信息学分析推测PcWRKY71编码蛋白分子量为37.41 kDa,理论等电点为6.72,属于不稳定的亲水性蛋白,定位于细胞核上,与甜樱桃和樱花的亲缘关系最近。荧光定量分析结果表明,PcWRKY71基因表达受干旱、盐害和低温胁迫诱导及激素SA和MeJA影响,说明PcWRKY71转录因子对甜樱桃砧木应答非生物胁迫和激素处理起着重要作用。In order to study the function of WRKY71 transcription factor and its response to abiotic stress and hormone treatment, the PcWRKY71 gene was cloned from sweet cherry rootstock ‘Gisela 6’. Sequence analysis showed that the complete open reading frame of PcWRKY71 was 1 014 bp, encoding 337 amino acids. It contained one WRKY conserved domain and one C_(2)H_(2)zinc finger domain, and belonged to the group Ⅱ of WRKY gene family. Bioinformatics analysis suggested that the molecular weight of PcWRKY71 protein was 37.41 kDa, and the theoretical isoelectric point was 6.72. It was an unstable hydrophilic protein, located on the nucleus, and had a close genetic relationship with Prunus avium and Prunus yedoensis. The qRT-PCR analysis showed that the expression of PcWRKY71 gene was induced by drought, salt stress and low temperature, as well as affected by SA and MeJA, which suggested that PcWRKY71 transcription factor played an important role in abiotic stress and hormone treatment responses.
