摘  要:目的探讨动脉粥样硬化模型小鼠外周血和主动脉组织中CCR2 mRNA表达变化趋势。方法将40只apoE基因敲除小鼠分为高脂饮食组和普通饮食组,每组20只,在喂养的第4周开始进行外周血和主动脉CCR2 mRNA测定,了解动脉粥样硬化发展过程中CCR2 mRNA表达的变化。在第2周和第14周对小鼠进行血脂测定,并对主动脉组织进行油红O染色以及免疫荧光检测验证模型建立并了解CCR2 mRNA在组织中的表达。结果高脂饮食喂养的apoE基因敲除小鼠外周血CCR2 mRNA在第4周即开始出现,在第10周达高峰并且维持较高水平,而主动脉组织中CCR2 mRNA的表达在第8周开始逐渐增加。高脂饮食组小鼠在第14周可见血脂显著增高,油红O染色提示主动脉粥样斑块形成,动脉粥样硬化模型建立成功,斑块内可见CCR2 mRNA大量表达。结论外周血CCR2 mRNA的升高可能是动脉粥样硬化相关早期单核细胞活化的一个有效预测指标。Objective To explore the changes of CCR2 mRNA expression in peripheral blood and aorta tissue in atherosclerosis model mice.Methods Forty apoE knockout mice(apoE-/-)were divided into high fat diet group and normal diet group,with twenty animals in each group.CCR2 mRNA expressions in aorta tissue and peripheral blood were measured every two weeks from week 4 of feeding to investigate CCR2 expression changes.Blood lipids in week 2 and week 14 were also detected.Oil O staining and immunofluorescence detection were used for the determination of atherosclerosis model establishment and CCR2 expressions in the plaque in week 14.Results CCR2 mRNA expression in peripheral blood began to increase from week 4 and peaked at week 10 from when it maintained a high level.While CCR2 mRNA expression in aorta tissue was increased from week 8.Blood hyperlipidemia,atherosclerotic plaque formation,successful establishment of atherosclerosis model,and high expression of CCR2 mRNA in plaque were observed at week 14 in high fat diet group.Conclusion Increased CCR2 mRNA in peripheral blood might be a useful marker for the early prediction of atherosclerosis associated monocytes activation.
