摘 要:目的观察清营生脉汤对心肌缺血再灌注损伤(MIRI)大鼠炎症因子的影响。方法将60只SD大鼠随机分为假手术组、模型组、阳性药组和清营生脉汤高、中、低剂量组,每组10只。灌胃14 d后,结扎大鼠心脏左冠状动脉前降支后再复灌建立心肌缺血再灌注模型。应用TTC染色法检测心肌梗死面积;ELISA法检测肌酸激酶(CK)、肌酸激酶同工酶(CK-MB)、白细胞介素-2(IL-2)、白细胞介素-4(IL-4)水平;Western blotting法检测JNK、P-JNK蛋白表达。结果与假手术组比较,模型组心肌梗死面积增加;与模型组比较,清营生脉汤高、中、低剂量组与阳性药组的心肌梗死面积明显减少;与模型组比较,清营生脉汤高、中、低剂量组CK和CK-MB含量显著降低(P<0.01),IL-2水平显著降低(P<0.01),IL-4水平显著升高(P<0.01),JNK蛋白表达无明显改变(P>0.05),P-JNK蛋白表达显著降低(P<0.01)。结论清营生脉汤可减轻MIRI大鼠心肌损伤,缩小心肌梗死面积,发挥心肌保护作用,并推测其减轻MIRI炎症损伤的作用可能与抑制JNK通路激活相关。Objective:To study the effect of Qingying Shengmai Decoction on the inflammatory factor of myocardial ischemia-reperfusion injury(MIRI)rats.Methods:A total of 60 SD rats were divided into the sham operation group,model group,positive drug group,Qingying Shengmai Decoction high-dose,medium-dose and low-dose groups,10 rats in each group.After 14 days of gavage,the left anterior descending branch of the left coronary artery of the rat was ligated and reperfusion to establish a MIRI model.The area of myocardial infarction was detected by TTC staining.The levels of CK,CK-MB,IL-2 and IL-4 were detected by ELISA.The protein expression of JNK and P-JNK was detected by Western blotting.Results:Compared with the sham operation group,the myocardial infarction area was increased and apoptosis was obvious in the model group.Compared with model group,the myocardial infarction area in Qingying Shengmai Decoction high-dose,medium-dose and low-dose groups and positive drug group was significantly reduced;compared with model group,the concentration of CK and CK-MB in Qingying Shengmai Decoction high-dose,medium-dose and low-dose groups was significantly decreased(P<0.01),the level of IL-2 was significantly decreased(P<0.01),the level of IL-4 was significantly increased(P<0.01),the expression of JNK protein showed no obvious changes,and the expression of P-JNK protein was significantly decreased(P<0.01).Conclusion:Qingying Shengmai Decoction can reduce myocardial injury in MIRI rats,reduce the area of myocardial infarction,and play a protective role on the myocardium.It is speculated that the reduction of MIRI inflammation may be related to the inhibition of JNK pathway.
