摘 要:为了探究不同比例的α-生育酚及α-生育三烯酚抗氧化能力的大小,共开展2个试验。试验一∶设置4个不同比例的α-生育酚及α-生育三烯酚,分别为α-生育酚(TE组)、α-生育酚∶α-生育三烯酚=6.5∶1(TT1组)、α-生育酚∶α-生育三烯酚=2.75∶1(TT2组)、α-生育酚∶α-生育三烯酚=1.5∶1(TT3组),生育酚及生育三烯酚之和均为100μmol/L,测定其羟自由基清除率、超氧阴离子清除率。试验二∶选用与试验一相同比例的α-生育酚及α-生育三烯酚,与猪小肠上皮细胞(IPEC-J2)共培养6 h后加入敌草快(Diquat,DQ)处理4 h,用CCK-8法测定细胞活力,用流式细胞仪测定细胞内活性氧含量。试验结果表明∶不同比例的α-生育酚与α-生育三烯酚对羟自由基的清除率存在差异,其中TT3组羟自由基清除率显著高于TE组(P<0.05),其余各组间无显著性差异;超氧阴离子清除率也受α-生育酚与α-生育三烯酚比例的影响,TT1组超氧阴离子清除率显著高于TE组(P<0.05),TT2、TT3组极显著高于TE组(P<0.01)。细胞培养试验表明∶敌草快处理后极显著降低了细胞活力(P<0.001),TT2预处理组细胞活力显著高于敌草快处理组(P<0.05),TT3预处理组细胞活力极显著(P<0.01)高于敌草快处理组,TT3组抵抗敌草快氧化应激的效果最佳;不同比例的α-生育酚与α-生育三烯酚对细胞内活性氧的含量也有显著的影响,与Ctrl组相比,加入敌草快后,各试验组细胞内活性氧含量极显著上升(P<0.01),与DQ组相比,TE+DQ、TT1+DQ、TT2+DQ组细胞内活性氧含量均显著下降(P<0.05),TT3+DQ组细胞内活性氧含量极显著下降(P<0.01),与TE+DQ组相比,TT2+DQ、TT3+DQ组细胞内活性氧含量显著下降(P<0.05)。因此∶不同比例的α-生育酚及α-生育三烯酚的抗氧化能力不同,其中α-生育酚∶α-生育三烯酚=1.5∶1时,其羟自由基清除率、超氧阴离子清除率最高;对IPEC-J2细胞的预保护作用最好,抗氧化能力最佳。In order to investigate the antioxidant capacity ofα-tocopherol andα-tocotrienol in different proportions,two experiments were conducted.In experiment 1,four different proportions ofα-tocopherol andα-tocotrienol were set up,respectively:α-tocopherol(TE group),α-tocopherol∶α-tocotrienol=6.5∶1(TT1 group),α-tocopherol∶α-tocotrienol=2.75∶1(TT2 group),α-tocopherol∶α-tocotrienol=1.5∶1(TT3 group),the sum of tocopherol and tocotrienol were 100μmol/L,and the scavenging rates of hydroxyl radical and superoxide anion were measured.In experiment 2,α-tocopherol andα-tocotrienol,the same proportions as in experiment 1,were co-cultured with porcine small intestine epithelial cells(IPEC-J2)for 6h and treated with diquat(DQ)for 4h.Cell viability was determined by CCK-8 method and intracellular reactive oxygen species content was determined by flow cytometry.The results showed that the hydroxyl radical scavenging rates ofα-tocopherol andα-tocotrienol were different,and the hydroxyl radical scavenging rates of TT3 group were significantly higher than those of TE group(P<0.05),but there was no significant difference among other groups.The superoxide anion clearance rate was also affected by the ratio ofα-tocopherol toα-tocotrienol.The superoxide anion clearance rate of TT1 group was significantly higher than that of TE group(P<0.05),and that of TT2 and TT3 groups was significantly higher than that of TE group(P<0.01).The cell culture test showed that the cell viability was significantly decreased(P<0.001)after treatment with diquat,the cell vitality of the TT2 group was significantly higher than that of DQ group(P<0.05),and the cell vitality of the TT3 group was significantly higher than that of DQ group(P<0.001).TT3 group had the best resistance to the oxidative stress of diquat.Different proportions ofα-tocopherol andα-tocotrienol also had significant effects on the content of intracellular reactive oxygen species.Compared with control group,the content of intracellular reactive oxygen species in ex
