摘 要:目的研究皮内针预处理对脑缺血再灌注(CI/R)大鼠神经功能的保护作用。方法将8周龄,体重为(230±10)g的SD清洁级雄性大鼠120只采用随机数字表法分为四组(假手术组、模型组、电针组和皮内针组),每组30只。皮内针组和电针组,分别以皮内针和电针对大鼠进行预处理后,制备大鼠脑中动脉阻塞(MCAO)再灌注模型,模型组仅接受造模处理,假手术组仅进行开颈手术,但不进行结扎。采用改良神经功能缺损评分(mNSS)分析造模后大鼠的神经功能受损程度,酶联免疫吸附试验法检测炎症因子白细胞介素(IL)-1β和IL-6含量,Western blot法检测凋亡相关蛋白(Caspase-3、Bax、BCL-2)的表达水平。结果模型组mNSS评分高于假手术组,差异有统计学意义(P<0.05),电针组和皮内针组mNSS评分低于模型组,差异有统计学意义(P<0.05)。模型组IL-1β和IL-6含量高于假手术组,差异有统计学意义(P<0.05)。电针组和皮内针组IL-1β和IL-6含量低于模型组,差异有统计学意义(P<0.05)。模型组病灶部位Caspase-3、Bax蛋白水平高于假手术组,BCL-2蛋白水平低于假手术组,差异有统计学意义(P<0.05),电针组和皮内针组Caspase-3、Bax蛋白水平低于模型组,BCL-2蛋白水平高于模型组,差异有统计学意义(P<0.05)。结论皮内针预处理可显著降低CI/R后大鼠病灶部位炎症因子,抑制神经细胞凋亡,减轻CI/R后大鼠脑神经功能损伤。此外,皮内针预处理具有方便,医疗成本较低的优势,有利于临床推广。To study the protective effect of intracutaneous acupuncture preconditioning on neural function in rats with cerebral ischemia/reperfusion(CI/R).Method A total of 120 SD male rats aged eight weeks and weighing(230±10)g were divided into four groups(sham operation group,model group,electro-acupuncture group and endothelium acupuncture group),with 30 rats in each group.In the endothelium acupuncture group and electro-acupuncture group,the rats were pretreated with endothelium acupuncture and electro-acupuncture respectively,and then the middle cerebral artery occlusion(MCAO)reperfusion model of rats was prepared.The model group only accepts modeling treatment.In the sham operation group,only open neck surgery was performed,but no ligation was performed.Neurological impairment was analyzed by modified neurological deficit score(mNSS),and the levels of inflammatory cytokines interleukin(IL)-1βand IL-6 were measured by enzyme-linked immunosorbent assay.The expression levels of Caspase-3,Bax and BCL-2 were detected by Western blot.Results mNSS score in the model group was higher than that in the sham operation group(P<0.05),mNSS scores in the electro-acupuncture group and endothelium acupuncture group were lower than that in the model group,and the differences were statistically significant(P<0.05).The contents of IL-1βand IL-6 in model group were higher than those in sham operation group,and the differences were statistically significant(P<0.05).The contents of IL-1βand IL-6 in electro-acupuncture group and endothelium acupuncture group were lower than those in model group,and the differences were statistically significant(P<0.05).The protein levels of Caspase-3 and Bax in model group were higher than those in sham operation group,and the protein levels of BCL-2 were lower than those in sham operation group,and the differences were statistically significant(P<0.05).The protein levels of Caspase-3 and Bax in electro-acupuncture group and endothelium acupuncture group were lower than those in model group,and the pro
