摘　　要：目的:研究JAK3抑制剂对肺纤维化大鼠模型外周血白细胞计数、淋巴细胞百分比、支气管肺泡灌洗液淋巴细胞百分比、肺组织病理的影响。方法:选取雄性SPF级Sprague dawley (SD)大鼠60只,根据随机数字表法分为5组,分别为正常对照组、模型对照组、糖皮质激素(GC)组、JAK3抑制剂组、JAK3抑制剂+糖皮质激素联合组,每组12只。使用博来霉素气管注射对除正常对照组以外的其他各组大鼠造模,造模6天后开始给各组大鼠灌胃相应药物或生理盐水,造模30天后收集样本,观察大鼠的一般情况,检测大鼠外周血淋巴细胞百分比、肺泡灌洗液淋巴细胞百分比、肺组织HE染色。结果:在外周血白细胞计数方面,各组大鼠差异无统计学意义(P>0.05),糖皮质激素组和JAK3抑制剂组与对照相比有所降低,而JAK3抑制剂联合糖皮质激素组白细胞计数高于对照组;在淋巴细胞方面,各组大鼠之间差异有统计学差异(P<0.05),其中JAK3抑制剂组与正常对照组和糖皮质激素组相比,差异有统计学意义(P<0.05),与模型对照组检测结果相似,而JAK3抑制剂联合糖皮质激素时淋巴细胞百分比的却出现了明显的降低,与模型对照组具有显著的统计学差异。在肺泡灌洗液中,各组大鼠差异无统计学意义(P>0.05),依然可以观察到JAK3抑制剂+糖皮质激素联合组的淋巴细胞百分比的降低,相比模型对照组明显减少;但与外周血不同的是,在支气管肺泡灌洗液中JAK3抑制剂组的淋巴细胞百分比少于正常对照组、糖皮质激素组。在肺组织病理he染色方面,与正常对照组相比,模型对照组肺组织有广泛性轻到中度的肺泡壁增厚、局部重度的肺泡壁增厚,伴有纤维组织增生;糖皮质激素组小血管和毛细血管淤血,伴有纤维素渗出和细胞浸润,肺组织结构完整;JAK3抑制剂组有较大范围的实质病变,肺泡壁毛细血管淤血,组织结构模糊;JAK3抑制剂+糖皮质激Objective:To study the effects of JAK3 inhibitors on peripheral blood leukocyte count, lymphocyte percentage,bronchoalveolar lavage fluid lymphocyte percentage, and lung histopathology in a rat model of pulmonary fibrosis.Methods:60male SPF-grade Sprague dawley(SD) rats were selected and divided into normal control group, model control group, glucocorticoid(GC) group, JAK3 inhibitor group and JAK3 inhibitor + glucocorticoid combination group according to the random number table method, with 12 rats in each group. Rats in all groups except the normal control group were molded using bleomycin tracheal injection and gavage of the appropriate drug or saline was started for each group 6 d after modeling, and samples were collected 30d after modeling. The general condition of rats was observed, and the percentage of peripheral blood lymphocytes, percentage of alveolar lavage fluid lymphocytes and HE staining of lung tissue were detected.Results:The differences in peripheral blood leukocyte counts of rats in each group were not statistically significant(P>0.05). The glucocorticoid and JAK3 inhibitor groups were reduced compared with the control, while the white blood cell count was higher in the JAK3 inhibitor combined with glucocorticoid group than in the control group, and in terms of lymphocytes, the differences between the groups of rats were statistically different(P<0.05). The JAK3 inhibitor group showed statistically significant differences compared with the normal control group and the glucocorticoid group(P<0.05), and the results were similar to those of the model control group, whereas the percentage of lymphocytes showed a significant decrease when the JAK3 inhibitor was combined with glucocorticoid, with statistically significant differences from the model control group. There was no statistically significant difference in alveolar lavage fluid between the groups(P>0.05), and still a decrease in the percentage of lymphocytes in the combined JAK3 inhibitor +glucocorticoid group could be observed, which was si
