摘 要:目的:探讨经典瞬时感受器电位通道(TRPC)对内皮细胞和平滑肌细胞增殖所引起的阻塞性病变的影响。方法:选取人体尸检标本的颈动脉中明显有动脉粥样硬化的组织进行处理后经染色检测其TRPC6蛋白表达情况。取小鼠饲养高脂饲料12周后建模对比常规喂养组的TRPC6斑块表达情况。基因敲除后,动脉动脉粥样硬化斑块中的平滑肌细胞及胶原纤维含量情况、血管壁增厚及血流阻力情况。结果:TRPC6在人体动脉粥样硬化斑块中的表达显著更高,经免疫组织化学(IHC)染色其平均光密度显著更高。小鼠模型组(0.004±0.010)高于对照组(0.063±0.012),差异有统计学意义(P<0.05)。基因敲除后,模型小鼠的动脉粥样硬化斑块中的巨噬细胞含量(16.55±1.12)显著少于之前的(28.21±2.23),差异有统计学意义(P<0.05);平滑肌细胞(27.26±2.51)显著高于之前的(20.92±1.61),差异有统计学意义(P<0.05);而胶原纤维差异不明显。基因敲除后,能明显改善小鼠的血管壁增厚及血流阻力情况。结论:TRPC6在动脉粥样硬化斑块中的表达显著调高,提示其参与了动脉粥样硬化斑块的发生及发展过程。Objective:To investigate the effect of classical transient receptor potential channels(TRPC) on obstructive lesions caused by proliferation of endothelial cells and smooth muscle cells.Methods:Tissues with apparent atherosclerosis in the carotid arteries of human autopsy specimens were taken for processing and stained to detect their TRPC6 protein expression. Mice were taken and modeled after 12 weeks of feeding high-fat chow, and TRPC6 plaque expression in the conventionally fed group was compared. After gene knockdown, the smooth muscle cell and collagen fiber content in the atherosclerotic plaque, vessel wall thickening and blood flow resistance were observed.Results:TRPC6 expression was significantly higher in human atherosclerotic plaques,and its mean optical density was significantly higher by immuno histochemistry(IHC) staining. It was(0.004 ± 0.010) in the mouse model group, which was higher than(0.063±0.012) in the control group, and the difference was statistically significant(P<0.05). After gene knockdown, the content of macrophages in atherosclerotic plaques of model mice was(16.55±1.12), significantly less than before(28.21±2.23), with statistically significant differences(P<0.05). Smooth muscle cells were(27.26±2.51), significantly higher than the previous(20.92±1.61), with a statistically significant difference(P<0.05), while the difference in collagen fibers was not significant. The gene knockout significantly improved blood vessel wall thickening and blood flow resistance in mice.Conclusion:The expression of TRPC6 was significantly elevated in atherosclerotic plaques, suggesting its involvement in the development and progression of atherosclerotic plaques.
