灵龟八法按时开穴灸对衰老大鼠睾丸细胞凋亡的影响

摘　　要：目的:观察灵龟八法按时开穴灸对衰老大鼠超氧化物歧化酶(SOD)、丙二醛(MDA)含量及抗凋亡调控因子B淋巴细胞瘤2(Bcl-2)和促凋亡调控因子Bcl相关X蛋白(Bax)表达的影响,探讨其作用机制。方法:SD大鼠随机分为空白组、模型组、灵龟八法组,每组8只。除空白组外,其余两组均以D-半乳糖溶液(500 mg/kg)腹腔注射复制亚急性衰老模型。造模成功后,灵龟八法组大鼠予以灵龟八法按时开穴灸治疗,每穴艾灸3壮,1次/d,连续28 d。采用ELISA法观察大鼠血清中SOD、MDA的含量;HE染色法观察大鼠睾丸组织一般形态和间质细胞数量的变化;TUNEL染色法检测大鼠睾丸细胞凋亡率;Western blot法检测大鼠睾丸组织中Bcl-2、Bax蛋白的表达水平。结果:与空白组比较,模型组大鼠血清SOD含量、睾丸组织间质细胞数量减少(P<0.01),血清MDA含量增多(P<0.01),睾丸细胞凋亡率、睾丸组织中Bax蛋白的相对表达量升高(P<0.01),睾丸组织中Bcl-2蛋白的相对表达量降低(P<0.01)。与模型组比较,灵龟八法组大鼠血清SOD含量、睾丸组织间质细胞数量增多(P<0.01),血清MDA含量减少(P<0.01),睾丸细胞凋亡率、睾丸组织中Bax蛋白的相对表达量降低(P<0.01),睾丸组织中Bcl-2蛋白的相对表达量升高(P<0.01)。结论:灵龟八法按时开穴灸能够起到延缓大鼠睾丸衰老的作用,其机制可能与平衡自由基的代谢,减少机体氧化损伤,进而抑制睾丸细胞凋亡有关。Objective To observe the effect of“lingguibafa”moxibustion at“opening”time on the level of superoxide dismutase(SOD),malondialdehyde(MDA),the protein expressions of B-cell lymphoma-2(Bcl-2)and Bcl-2 associated X protein(Bax)in aging model rats.Methods SD rats were randomly divided into blank control,model and lingguibafa groups,with 8 rats in each group.The aging model was established by daily intraperitoneal injection of D-galactose(500 mg/kg)for 42 consecutive days.After successful modeling,moxibustion intervention was applied at“lingguibafa”acupoints,3 moxa-cone for each acupoint,once a day for 28 consecutive days.The contents of SOD and MDA in serum were detected by ELISA.Morphological changes of testicular tissue and the number of Leydig cells were observed after HE staining.Apoptosis rate of testicular cells was detected by TUNEL staining.The protein expression levels of Bcl-2 and Bax in testis were detected by Western blot.Results Compared with the blank control group,the SOD content in the serum,the number of testicular Leydig cells,the expression level of Bcl-2 protein in testis were significantly decreased(P<0.01)in the model group,while the MDA content in the serum,the apoptosis rate of testicular cells and the expression level of Bax protein in testis were significantly increased(P<0.01).Compared with the model group,the SOD content in serum,the number of testicular Leydig cells,the Bcl-2 protein expression in testis were significantly increased(P<0.01),while the MDA content in serum,the apoptosis rate of testicular cells,the expression level of Bax protein in testis were significantly decreased(P<0.01)in the lingguibafa group.Conclusion Moxibustion on acupoints at“opening”time can delay testicular aging,and the mechanism may be related to balancing the metabolism of free radicals,reducing oxidative damage,and inhibiting the apoptosis of testicular cells.
