摘  要:目的了解红河州HIV/HCV合并感染者中HCV的基因型分布。方法收集2018-2019年红河州HIV/HCV抗体阳性的血浆246份,进行HCV病毒载量检测,对有病毒载量的样品E1E2区和N55B区采用巢氏PCR扩增,扩增产物经基因序列测定,所得序列通过构建系统进化树确定HCV的分子亚型。结果结合2个基因片段,最终120份样品获得了分型结果。3b为主要亚型,占30.8%(37/120),其他亚型按照比例依次为3a(21.7%,26/120)、6a(17.5%,21/120)、6n(16.7%,20/120)、其他亚型(13.3%,16/120)。各HCV亚型在民族分布上的差异有统计学意义(P<0.05)。分别计算各亚型在E1E2区和NS5B区的基因距离,结果显示在3a、3b、6a、1b、6n这5种亚型中,3a的基因距离最大,且3a、6a、1b、6n的基因距离均与3b的基因距离有显著性差异(P<0.05)。结论在红河州HIV/HCV合并感染者中,3b是主要流行亚型,且在该人群中有较长的流行时间。Objective To investigate the distribution of hepatitis C virus(HCV)genotypes among HIV/HCV coinfected individuals in Honghe prefecture.Methods 246 HIV/HCV positive plasma samples were collected from Honghe during 2018-2019.All the samples were tested for HCV viral load.E1E2 and NS5B genes were amplified using nested-polymerase chain reaction from the samples with detected viral load.The HCV subtypes were determined through phylogenetic analysis after sequencing.Results Based on phylogenetic analysis of E1E2 and NS5B gene fragments,a total of 120 samples were genotyped.3b was the predominant subtype(30.8%,37/120),followed by 3a(21.7%,26/120),6a(17.5%,21/120),6n(16.7%,20/120)and others(13.3%,16/120).3b had the most significant genetic distance,and the gene distance of 3a,6a,1b,and 6n was significantly different from that of 3b(P<0.05).Conclusions HCV subtypes 3b was predominant among HIV/HCV co-infected individuals in Honghe,which has long prevalence duration in this population.
