摘  要:为探讨限食延长寿命的分子机制,以家蚕品种秋丰为研究对象,从2龄幼虫起限制采食(DR),每日1次给桑,自由采食16 h后强制停食8 h;以每日2次给桑的全天自由采食(AL)为对照。4-5龄期雌、雄分开饲养,选取3个与无脊椎动物寿命相关基因的家蚕同源基因BmSir2(Bombyx mori silencing information regulator 2)、BmAmpk(Bombyx mori adenosine activated protein kinase)和BmCox(Bombyx mori cytochrome C oxidase),利用qRT-PCR技术分析5龄3 d时的mRNA水平。进一步用RNAi技术敲减上述3个基因,调查其对成虫寿命的影响。结果显示,家蚕幼虫各组织中,3个寿命相关基因在DR组与AL组均呈现不同程度的表达差异;与雌性相比,限食后雄性的BmSir2、BmAmpk、BmCox在各组织中的表达上调更加明显;RNAi后3个基因的表达均显著下调,与对照组(dsGFP)相比成虫寿命显著缩短。结果表明限食促进寿命相关基因表达上调,且促进寿命延长。To investigate the molecular mechanism of dietary restriction in prolonging life-span,Qiufeng,a Bombyx mori bivoltine strain,was used as the research object.Dietary restriction(DR)groups of the 2 nd instar larvae were given mulberry leaves once a day,and then forced to fast for 8 h after being fed freely for 16 h.As a control,ad libitum feeding(AL)was given to mulberry twice a day with free feeding all day.The female and male larvae were reared separately from the 4 th to 5 th instars.The mRNA levels of BmSir2(Bombyx mori silencing information regulator 2),BmAmpk(Bombyx mori adenosine activated protein kinase)and BmCox(Bombyx mori cytochrome C oxidase),the silkworm homologs of three invertebrate life-span-related genes,were analyzed by qRT-PCR technology at the 5 th instar day-3 larvae.The above three genes were further knocked down by RNA interference(RNAi)technology to investigate their effects on adult lifespan.The results showed that the expression of three life-span related genes revealed differences in some way between the DR and the AL groups.BmSir2,BmAmpk,and BmCox genes were more significantly up-regulated in the tissues of male larvae in DR group compared with female.All three genes were significantly down-regulated after RNAi,and the adult life-spans were significantly shortened compared with the control group(dsGFP).These results indicate that dietary restriction up-regulates the expression of life-span related genes,and promotes adult life-span.
