摘　　要：目的 应用串联质谱技术鉴定重组人乳头瘤病毒L1蛋白C端的氨基酸序列。方法 将C端富含精氨酸和赖氨酸的重组人乳头瘤病毒L1蛋白酶解成合适长度的肽段混合物,运用液质联用技术进行鉴定。结果 分别对6、11、16、18亚型的重组人乳头瘤病毒L1蛋白的C端氨基酸序列进行了分析确证,均与理论序列一致。结论 所用分析方法简单可行,为HPV疫苗的结构表征和质量控制提供了参考和依据。OBJECTIVE To identify C-terminal sequence in recombinant human papillomavirus capsid protein L1 by LC-MS/MS.METHODS The peptides with suitable length were analyzed using LC-MS/MS after they with arginine and lysine rich in C-terminal were digested by trypsin directly.RESULTS The C-terminal sequence of type 6,11,16 and 18 recombinant human papillomavirus capsid protein L1 were identified, which were identical with the theoretical sequence completely.CONCLUSION The identification of C-terminal sequence method is applicable, which can provide evidence for the structure characterization and quality control of HPV vaccine.
