摘 要:目的 优化超临界CO_(2)萃取法萃取南方红豆杉中挥发油和10-DABⅢ的工艺条件。方法 以萃取时间、萃取温度、萃取压力、分离温度、物料与夹带剂的比例和夹带剂浸泡时间等为考察因素,以红豆杉中挥发油的得率和10-DABⅢ的含量为评价指标,采用正交试验优选最佳工艺条件。结果 最佳工艺条件为以无水乙醇为夹带剂,物料与夹带剂比例为1:3,萃取压力270 bar、萃取温度35℃、分离温度40℃,萃取时间2.5 h。红豆杉挥发油的得率为23.422%,10-DABⅢ的含量0.546‰。结论 所用工艺稳定、可行,可用于红豆杉中挥发油和10-DABⅢ的提取。OBJECTIVE To optimize the supercritical CO_(2)fluids extraction(SFE) method to extract the volatile oil and 10-deacetyl baccatin(10-DABⅢ) from Taxus chinensis.METHODS The extraction technology of volatile oil and 10-DABⅢ from Taxus chinensis was optimized by orthogonal experiment using extraction time, extraction temperature, extraction pressure, separation temperature, material-entrainer ratio, entrainer soak time as factors and with the contents of volatile oil and 10-DABⅢ.RESULTS The optimal technology was as follows: absolute ethanol as entrainer, with the entrainer-materials ratio of 3:1,extraction pressure of 270 bar, extraction temperature of 35 ℃,separation temperature of 40 ℃,and extraction time of 2.5 h.The yield of volatile oil was 23.422%,and the content of 10-DABⅢ was 0.545‰.CONCLUSION The process is stable and feasible for the extraction of volatile oil and 10-deacetyl baccatin from T.chinensis.
