摘 要:目的 考察瘤果黑种草子总皂苷(TSSN)的抗炎和免疫调节作用。方法 建立脂多糖(LPS)诱导的小鼠单核巨噬细胞白血病细胞RAW264.7炎症模型,采用Griess法检测NO分泌水平,ELISA法检测细胞上清TNF-α、IL-6、IL-1β含量,采用qRT-PCR方法考察TSSN对NF-κB信号通路相关靶点mRNA表达水平的影响;酶动力学实验检测总皂苷对环氧酶-2(COX-2)以及5-脂氧酶(5-LO)的抑制活性;通过MTS法及ELISA法检测TSSN对静息期淋巴细胞的影响,并以TSSN对刀豆蛋白(ConA)、LPS分别诱导的小鼠T、B淋巴细胞增殖的影响,以及对T淋巴细胞分泌细胞因子TNF-α、IL-2、IL-6、IFN-γ的影响来考察TSSN的免疫调节作用。结果 在LPS诱导的RAW 264.7炎症模型中,TSSN能显著降低LPS诱导的RAW264.7分泌炎症介质NO和促炎症因子TNF-α、IL-6、IL-1β水平,显著抑制TNF-α、IL-6、IL-1β m RNA表达水平,抑制iNOS、NF-κB p65 mRNA表达水平;在12.5~200μg·ml-1剂量时能显著抑制COX-2活性,在1 000~4 000μg·ml^(-1)剂量时对5-LO也具有显著抑制作用;此外,TSSN对ConA、LPS分别诱导的小鼠T、B淋巴细胞增殖具有显著抑制作用,明显抑制T淋巴细胞分泌TNF-α、IL-2、IL-6及IFN-γ。结论 TSSN具有较好的抗炎活性,其机制可能与抑制NF-κB信号通路有关。此外,TSSN具有较好的免疫调节作用,其机制有待进一步研究。This study was designed to investigate the anti-inflammatory and immunomodulatory effects of total saponins from Seeds of Herba nigra(TSSN). The model of RAW264.7 inflammation was established with lipopolysaccharide(LPS) induction, and the level of NO secretion was detected by Griess method. The contents of TNF-α, IL-6 and IL-1β in supernatant were detected by ELISA;the effects of TSSN on TNF-α, IL-6, IL-1β,iNOS, NF-κB p65 mRNA expression were investigated by qRT-PCR;the inhibitory activities of TSSN against cyclooxygenase-2(COX-2) and 5-Lipoxygenase(5-LO) were determined by enzymatic assay. Furthermore, MTS and ELISA methods were used to determine the effects of TSSN on resting lymphocytes, the proliferations of T and B lymphocytes induced by ConA and LPS, respectively, and the secretions of IL-2, IL-6 and IFN-γ in T lymphocytes.Data showed that the levels of NO, TNF-α, IL-6 and IL-1β were increased in RAW 264.7 inflammatory cell model group. TSSN significantly inhibited the secretion of NO, TNF-α, IL-6, IL-1β and inhibited mRNA expression levels of TNF-α, IL-6, IL-1β, iNOS, NF-κB p65. In addition, at the dose of 12.5~200 μg/ml, TSSN showed significantly inhibitory activities on COX-2, and at the dose of 1000~4000 μg/ml, TSSN also inhibited the activity of 5-LO. In T and B lymphocyte proliferation tests, TSSN significantly inhibited the proliferation of T and B lymphocytes induced with ConA or LPS. TSSN also obviously inhibited the secretions of TNF-α, IL-2, IL-6 and IFN-γ. In summary, TSSN has a good anti-inflammatory activity, which may be related to the inhibition of NF-κB signaling pathway. In addition, TSSN demonstrated goodimmunomodulation effects, which needs further study.
