通窍鼻渊丸对过敏性鼻炎大鼠调节作用及机制研究

摘　　要：目的探讨通窍鼻渊丸对过敏性鼻炎(AR)大鼠的调节作用及机制。方法采用卵清白蛋白结合佐剂诱导AR大鼠模型;灌胃给药2周后进行行为学评价,采用病理学观察、蛋白免疫印迹、酶联免疫吸附进行生化及组织检测。结果灌胃给药后,AR大鼠行为显著改善;鼻黏膜结构逐渐完整,间隙水肿及炎性细胞浸润情况减轻;血清细胞因子白细胞介素-4、免疫球蛋白E质量浓度显著降低,白细胞介素-2、干扰素γ质量浓度显著升高;免疫组织化学TOLL样受体2、TOLL样受体4阳性率均降低;T细胞转录因子蛋白表达显著增强。结论通窍鼻渊丸可改善卵清白蛋白诱导的过敏性鼻炎相关症状,其作用可能与调控T细胞转录因子表达、调节Th1/Th2平衡有关。Objective To investigate the regulatory effect and mechanism of Tongqiao Biyuan pill(TQBYP)on rats with allergic rhinitis.Methods AR model was induced by ovalbumin(OVA)combined with adjuvant,and behavioral evaluation was performed 2 weeks after oral administration;pathological observation,Western blotting and enzyme-linked immunosorbent assay were used for biochemical and tissue detection.Results After administration,the results showed that the AR rats'behavior could be obviously improved,the nasal mucosa was gradually integrated,and the edema of clearance and infiltration of inflammatory cells were alleviated.The contents of interleukin-4 and immunoglobulin E were decreased while contents of interleukin-2 and interferon-γwere increased.The positive rate of Toll-like receptor 2 and Toll-like receptor 4 in immunohistochemistry was decreased and the expression of T-bet increased.Conclusion TQBYP can improve OVAinduced allergic rhinitis-related symptoms,and the effect may be related to the regulation of T-bet expression and th1/th2 balance.
