摘 要:目的以乙醛中毒果蝇为模型,筛选并优化能显著改善乙醛中毒果蝇生存率的肉苁蓉多糖提取工艺。方法肉苁蓉药材粗粉依次用乙酸乙酯和乙醇脱脂处理,加入木瓜蛋白酶、纤维素酶和果胶酶,采用L9(34)正交法和复合酶辅助超声法提取肉苁蓉多糖,采用乙醛中毒果蝇模型筛选出药效活性最佳的肉苁蓉多糖。结果肉苁蓉多糖最佳提取工艺为木瓜蛋白酶(2.00%)、纤维素酶(2.00%)、果胶酶(0.40%),m(料)∶m(水)=1∶25,温度54.1°C,pH 5.2,酶解2 h,单次提取时间45 min,提取次数3次。结论采用传统方法筛选肉苁蓉多糖提取工艺,组间无显著性差异。结合正交实验结果,进一步采用改善乙醛中毒果蝇生存率为药效指标,为多糖的提取提供科学、合理的理论依据。Objective To take acetaldehyde poisoned drosophila as a model for screening and optimizing the extraction process of polysaccharides from Cistanche deserticola,so that the optimum polysaccharide can significantly improve the survival rate of acetaldehyde poisoned drosophila.Methods The crude powder of Cistanche deserticola was successively extracted with ethyl acetate and ethanol.Papain,cellulase and pectinase were added into defatted Cistanche deserticola powder,and the polysaccharides were extracted via the L9(34)orthogonal method and enzymolysis combined with the ultrasonic method.The extraction process of polysaccharides from Cistanche deserticola was screened with the best pharmacodynamic activity by drosophila acetaldehyde poisoned mode.Results The optimal extraction conditions of Cistanche deserticola polysaccharide were papain(2.00%),cellulase(2.00%),pectinase(0.40%),solid-liquid ratio of 1∶25,temperature of 54.1°C,pH of 5.2,enzymatic hydrolysis of 2 hours,single extraction time of 45 minutes,and extraction times of 3.Conclusion There was no significant difference in the extraction process of polysaccharides by traditional methods.The survival rate of acetaldehyde poisoned drosophila was used as the index to provide a scientific and reasonable theoretical basis for the extraction of polysaccharide.
