摘 要:目的 观察高渗盐水(HS)对于大鼠脊髓损伤(SCI)模型神经功能的保护作用。方法 SD大鼠随机分为三组。sham组大鼠只行椎板切除术。SCI组大鼠SCI造模后不应用药物治疗。HS组大鼠SCI造模后静脉输注7.5%HS,5 mL/Kg·d。采用改良Allen′s方法在T10节段制作SCI动物模型,SCI后7 d,采用BBB运动功能评分评估大鼠神经功能恢复情况,然后处死大鼠取伤段脊髓组织,采用病理切片HE染色观察组织形态,采用Western Blot检测AQP4表达情况,并用相应试剂盒检测铁离子、ROS、IL-1β和TNF-α的组织内水平。结果 与sham组比较,SCI后,SCI组及HS组大鼠BBB评分明显降低,肢体运动功能出现障碍,脊髓组织出现水肿,组织内AQP4、铁离子、ROS、IL-1β及TNF-α水平升高。而HS组大鼠较SCI组BBB评分明显改善,病理结果显示HS组大鼠脊髓组织水肿程度低于SCI组,并且HS组损伤脊髓组织内AQP4、铁离子及ROS、IL-1β和TNF-α的水平明显低于SCI组。结论 HS能通过减轻脊髓水肿,降低ROS及炎性因子水平来减轻脊髓继发性损伤,改善SCI后大鼠肢体功能。Objective To observe the protective effect of hypertonic saline on the neurological function of rats with spinal cord injury(SCI).Methods Sprague-Dawley rats were randomly divided into three groups. The sham group was subjected to laminectomy. The SCI group was used to model spinal cord injury without drug treatment. The hypertonic saline group comprise rats with SCI infused with 7.5% hypertonic saline(5 mL/Kg·d). SCI model was induced at the T10 segment through the modified Allen′s method. The recovery of the rats′ neurological function was evaluated through the Basso-Beattie-Bresnahan(BBB) motor function score after 7 days SCI induction. The rats were then sacrificed, and the injured spinal cord tissues were collected. The organizational form of the pathological sections was observed by hematoxylin-eosin staining, and aquaporin-4(AQP4) expression was detected by Western Blot method. The levels of iron ions, reactive oxygen species(ROS), IL-1β, and TNF-α in tissues were detected using corresponding kits.Results Compared with the sham group, after spinal cord injury, the BBB score of the rats in the SCI group and the HS group was significantly reduced, the limb motor function was impaired, the spinal cord tissue was edema, and the levels of AQP4, iron ion, ROS, IL-1β and TNF-α in the tissue increased. Compared with the SCI group, the BBB score of the rats in the HS group was significantly improved. The pathological results showed that the degree of edema in the spinal cord tissue of the rats in the HS group was lower than that in the SCI group, and the level of AQP4, iron ions, ROS, IL-1β and TNF-α in the injured spinal cord tissue of the HS group were significantly lower than that in the SCI group.Conclusion Hypertonic saline can reduce spinal cord edema, reduce the levels of ROS and inflammatory factors, reduce secondary spinal cord injury, and improve limb function in rats after spinal cord injury.
