摘  要:目的:研究双叶清肺止咳糖浆的祛痰、抗炎作用及其作用机制。方法:通过大鼠玻璃毛细管排痰实验和小鼠气管酚红排泌实验,观察双叶清肺止咳糖浆的祛痰作用;通过小鼠耳肿胀实验观察双叶清肺止咳糖浆的抗炎作用,通过小剂量脂多糖(LPS)诱导革兰阴性菌感染,建立大鼠感染后支气管模型,用ELISA法检测空白组、双叶清肺止咳糖浆高、中、低剂量组、阳性对照组左肺BALF中P物质(SP)、降钙素基因相关肽(CGRP)、鼠神经生长因子(NGF)的含量。结果:(1)大鼠排痰实验中,双叶清肺止咳糖浆高、中剂量组、氯化铵组大鼠排痰量较空白组增加,差异显著(P<0.01);(2)小鼠气管酚红排泌实验中,双叶清肺止咳糖浆高剂量组、氯化铵组、急支糖浆组能升高排泌酚红浓度,与空白组比较,差异显著(P<0.05,P<0.01);(3)小鼠耳肿胀实验中,双叶清肺止咳糖浆高剂量组、醋酸泼尼松组、急支糖浆组耳肿胀度较空白组降低,差异显著(P<0.05,P<0.01);(4)模型实验中,与正常组比较,模型组SP、CGRP、NGF的含量明显升高,双叶清肺止咳糖浆高、中剂量组、美敏伪麻溶液组SP、CGRP、NGF的含量与模型组比较显著降低,差异具有统计学意义(P<0.05,P<0.01)。结论:双叶清肺止咳糖浆能促进排痰,减轻炎症反应,减少大鼠咳嗽次数,保护受损炎症组织,其机制可能与改善大鼠SP和CGRP、NCF表达有关。Objective:To study the expectorant and anti-inflammatory effects of shuangye qingfei cough syrup.Methods:The expectorant effect of shuangye qingfei cough syrup was observed by glass capillary excretion test in rats and tracheal phenol red excretion test in mice;the anti-inflammatory effect of shuangye qingfei cough syrup was observed by ear swelling test in mice, and the post-infection bronchial model was established in rats by inducing gram-negative bacteria infection with small doses of lipopolysaccharide(LPS),The contents of substance P(SP),calcitonin gene-related peptide(CGRP) and murine nerve growth factor(NGF) in the left lung BALF of the blank group, high, medium and low dose groups and positive control group were measured by ELISA.Results:In the rat sputum excretion test, the amount of sputum excreted in the high and medium dose groups of shuangye qingfei cough syrup and ammonium chloride group increased compared with the blank group, and the difference was significant(P<0.01);In the mouse tracheal phenol red excretion test, the high dose group of shuangye qingfei cough syrup, ammonium chloride group and acute syrup group could increase the concentration of excreted phenol red, and the difference was significant compared with the blank group(P<0.05,P<0.01);In the mouse ear swelling, in the model test, the number of SP,CGRP and NGF were significantly higher in the model group compared with the normal group, and the number of SP,CGRP and NGF were significantly lower in each dosing group compared with the model group(P<0.05,P<0.01).The differences were statistically significant(P<0.05,P<0.01).Conclusion:Shuangye qingfei cough syrup can promote sputum excretion, reduce the inflammatory response, and decrease the number of coughs in rats, and the mechanism may be related to the improvement of SP and CGRP,and NCF expression in rats.
