摘 要:目的探讨与分析CT胸部平扫及增强评价肺动脉高压(PAH)大鼠心脏功能的价值。方法将40只SPF級雄性Wistar大鼠按照随机数字表法平分为2组-对照组与模型组,模型组采用一次性腹腔注射1.0%野百合碱(50 mg/kg)建立肺动脉高压大鼠模型,对照组一次性注射0.9%氯化钠溶液(5.0 mL/kg)。采用CT胸部平扫及增强评价肺动脉高压大鼠心脏功能,采用蛋白质印迹法检测肺组织内皮型一氧化氮合酶(eNOS)与蛋白激酶G1(PKG-1)蛋白表达情况并进行相关性分析。结果模型组造模第4周与造模第8周的平均肺动脉压均显著高于对照组,差异均有统计学意义(P<0.05),模型组均造模成功。模型组造模第4周与造模第8周的CT胸部平扫肺动脉直径与右室横径均明显高于对照组,差异均有统计学意义(P<0.05)。模型组造模第4周与造模第8周的CT胸部增强三尖瓣反流速度、主肺动脉峰值流速、右肺动脉峰值流速明显高于对照组,差异均有统计学意义(P<0.05)。模型组造模第4周与造模第8周的肺组织eN0S、PKG-1相对表达水平明显高于对照组,差异均有统计学意义(P<0.05)。在模型组造模第8周中,Pearson线性相关分析显示,肺动脉直径、右室横径、三尖瓣反流速度、主肺动脉峰值流速、右肺动脉峰值流速与肺组织eNOS、PKG-1蛋白相对表达水平均呈正相关性(r=0.582、0.613、0.598、0.602.0.546;r=0.6820.589、0.613、0.578、0.581,P<0.05)。结论肺动脉高压大鼠多伴随有肺组织eN0S与PKG-1蛋白的高表达,CT胸部平扫及增强能有效评价肺动脉高压大鼠心脏功能,且CT胸部平扫及增强扫描指标与eNOS、PKG-1蛋白表达水平呈现相关性。Objective To explore and analysis the values of CT chest plain scan and enhancement in evaluating cardiac function in ms with pulmonary arterial bhypertension(PAH).Methods Forty male Wistar rats of SPF grade were divided into two groups according to random number table-control group and model group-The model group was given a one-time intraperitoneal injection of 1.0%monocrotaline(50 mg/kg)to establish a pulmonary bypetension rat model,and the control goup was given a one-time injection of normal saline(5.0 mL/kg).The cardiac function of ralts with pulnonary arterial bypertension were eraluated by CT chest plain scan and cubancement,and the expresion of eN05 and PKG-1 protein in lung tsse were detected by Westem bloting,and the crelation were analyoed.Results The mean pulmonary arterial pressure in the model group at the 4th week and the 8th week after modeling were higher than that in the control group,the dfferences were stais-tically signifcant(P<0.05),and the model group were scesfully established.The pulmonary antery diameter and right ventricular transverse dianeter of CT cheet plain scan in the model group at the 4th week and the 8th week after modeling were higher than those in the control group,the diferences were satistically signifcant(P<0.05).The CT chest-enhanced tricuspid rgugitation velocity,peak flow velocity of main pul-monary artery and peak flow velocity of night pulmonary artery in the model group were higher than thoee in the control group at the 4th week and the 8th weck ater modeling,the dferences were satistically sgnificant(P<0.05).The relotve expression levels of eN05 and PKG-1 in the lung tssue of the model group were higher at the 4th wock and the 8th wock after modeling were higher than the contml group,the differences were saistically significant(P<0.05).In the 8th week of modeling in the model group,Pearson linear crrelation analysis showed that the pulmonary artery diameter,right ventricular transverse diameter,ticuspid rgugitationo veboeity,peak flow velocity of main pulmonary art
