桂枝汤标准汤剂及配方颗粒中芍药苷、肉桂酸和甘草酸铵的含量对比研究

摘  要:目的采用高效液相色谱-二极管阵列检测法(HPLC-DAD)测定桂枝汤标准汤剂与桂枝汤配方颗粒中芍药苷、肉桂酸和甘草酸铵三种成分的含量并进行对比。方法采用JADE-PAK KPEC-C_(18)(250 mm×4.6 mm,5μm)色谱柱,流动相为乙腈-0.1%磷酸水溶液,梯度洗脱,柱温35℃,流速1.0 ml/min。结果芍药苷、肉桂酸和甘草酸铵分别在2.57~51.44μg/ml、2.00~40.19μg/ml、2.66~53.26μg/ml浓度范围内有良好的线性关系(r2值分别为0.9998、0.9997和0.9999),检测限(RSN=3)分别为0.086μg/ml、0.033μg/ml和0.177μg/ml;精密度RSD为0.65%、0.33%和0.41%;平均回收率为96.2%、98.6%和96.4%。结论桂枝汤标准汤剂中的芍药苷与肉桂酸含量高于配方颗粒,而甘草酸铵的含量低于配方颗粒。本研究能为中药煎煮工艺提供技术参考,为规范配方颗粒临床用药和统一配方颗粒质量提供理论依据。Objective A high performance liquid chromatography with diode array detection(HPLC-DAD)method was developed for the simultaneous determination of three compounds including paeonia sinensis,cinnamic acid and ammonium glycyrrhizinate and for comparing the content of above components of Guizhi Standard Decoction and Guizhi Formula Granules Decoction.Methods The samples were separated with an ODS-C_(18) column(250 mm×4.6 mm,5μm)by gradient elution using water containing 0.1%phosphoric acid and acetonitrile as mobile phase,with a flow rate of 1 ml/min at 35℃.Results The standard curves revealed a good linear relationship(r2=0.9998,0.9997 and 0.9999)in the ranges of 2.57~51.44μg/ml for paeonia sinensis;2.00~40.19μg/ml for cinnamic acid and 2.66~53.26μg/ml for ammonium glycyrrhizinate,respectively.The limits of detection were 0.086,0.033,and 0.177μg/ml(RSN=3),respectively.The RSD of precision were 0.65%,0.33%and 0.41%,respectively.The average recovery rates were 96.2%,98.6%and 96.4%,respectively.Conclusion The contents of peony glycoside and cinnamic acid in Guizhi Decoction are shown to be higher than those in Guizhi Formula Granules Decoction.This study provides a technical reference for the decoction of traditional Chinese medicine,and provides a theoretical basis for standardizing the clinical use of formula granules and the quality of unified formula granules.
