摘 要:为了明确发生于海南海口永兴镇的玉米小斑病的病原菌,本研究对玉米叶片发病部位进行了病原菌的分离鉴定。结果表明,8个病原菌分离株在PDA培养基上生长形态基本一致,通过对其中一个分离株HNBM011菌丝生长和孢子形态进行显微观察,以及ITS序列分析,发现该病原菌为玉蜀黍平脐蠕孢菌(Bipolaris maydis),与四川玉米小斑病病原菌分离株(Bipolaris maydis isolate)DY-5-2高度同源。回接实验进一步证实了HNBM011分离株为玉米小斑病病原菌。此外,通过对68种不同的玉米自交系接种HNBM011,筛选出一个高抗玉米自交系金博士825父本和一个高感自交系乐农79母本,这为将来克隆抗玉米小斑病基因,实现抗病育种奠定了基础。In this study,the pathogen of maize southern leaf blight disease was isolated and identified from the corn planting area in Yongxing Town,Haikou,Hainan Province.Eight fungi isolates with almost the same growth morphology on PDA medium were isolated from the leaf disease spots,and one isolate HNBM011 was selected for the microscopic observation of mycelial growth and spore morphology,as well as ITS sequencing.The results showed that the isolate HNBM011 was Bipolaris maydis with a high similarity to the Sichuan isolate DY-5-2.Back inoculation assay confirmed that HNBM011 was the pathogen of maize southern leaf blight disease.Furthermore,by screening of 68 different maize inbred lines inoculation with HNBM011,that the Gold Doctor 825 male parent displayed complete disease resistance against HNBM011,and Gold Doctor Lenong 79 female parent was full susceptible to HNBM011.Overall,these results lay a foundation for further cloning of the resistance genes and breeding resistant cultivars against maize southern leaf blight disease in the future.
