摘  要:目的:研究木香烃内酯对于心肌细胞氧化损伤的保护作用及其与凋亡相关通路的相关性。方法:培养H9C2心肌细胞,分3组,分别为:正常组(Control)、氧化损伤组(Oxidative Damage Group, ODG)、木香烃内酯组(Costunolide);其中氧化损伤组H9C2心肌细胞通过双氧水构建氧化损伤模型,木香烃内酯组在氧化损伤组的基础上使用木香烃内酯预处理;通过CCK8实验测定各组细胞活力值,流式细胞仪检测各组细胞培养基上清液氧化应激及抗氧化应激指标的表达,Western blot检测各组p53通路蛋白的表达。结果:与Control组相比,ODG组与Costunolide组细胞活力值均明显下降(均P<0.05),ROS、MDA表达量均明显上升(均P<0.05),SOD、GSH表达量均明显下降(均P<0.05);与ODG组相比,Costunolide预处理后,细胞活力值明显提升(P<0.05),ROS、MDA表达量均明显下降(均P<0.05),SOD、GSH表达量均明显上升(均P<0.05),Caspase-3、p53的表达量明显降低(均P<0.05),Bcl-2表达量明显上升(P<0.05)。结论:木香烃内酯预处理可有效发挥对双氧水致心肌细胞氧化损伤的保护作用,其机制可能与p53相关信号通路相关。Objective: To research the protective effect of Costunolide on H9C2 cardiomyocytes oxidative damage induced by hydrogen peroxide and its correlation with p53 signal pathway. Methods: H9C2 cardiomyocytes were cultured and divided into three groups: Control Group, Oxidative Damage Group(ODG) and Costunolide Group;H9C2 cardiomyocytes in oxidative damage group were cultured with hydrogen peroxide to establish oxidative damage model, and Costunolide group was pretreated with Costunolide on the basis of oxidative damage group;The optimal protective concentration of Costunolide against oxidative damage was determined by CCK8 experiment, the expression of oxidative stress and antioxidant stress in each group was detected by ROS detection kit, and the expression of p53 signal pathway related proteins in each group were detected by Western blot.Results: Compared with control group, the cell viability of ODG group and Costunolide group were lowered significantly(all P<0.05), the expression of ROS and MDA were increased significantly(all P<0.05), and the expression of SOD and GSH were decreased significantly(all P<0.05). Compared with ODG group, after Costunolide pretreated, the cell viability was increased significantly(P<0.05), the expression of ROS and MDA were decreased significantly(all P<0.05), the expression of SOD and GSH were increased significantly(all P<0.05), the expressions of Caspase-3 and p53 were decreased significantly(all P<0.05) and the expression of Bcl-2 was increased significantly(P<0.05). Conclusion: Costunolide pretreatment can effectively protected myocardial cells from oxidative damage induced by hydrogen peroxide, and its mechanism may be related to the activation of p53 signaling pathway.
