摘 要:目的:探究薯蓣皂苷对单侧输尿管梗阻(unilateral ureteral obstruction, UUO)模型小鼠肾组织病理学改变和TNF-α表达的影响。方法:C57BL/6N小鼠80只,随机分为假手术组、UUO模型组(UUO模型组3 d、UUO模型组7 d)、薯蓣皂苷低剂量(50 mg/kg)组(UUO+薯蓣皂苷3 d组、UUO+薯蓣皂苷7 d组)、薯蓣皂苷高剂量(100 mg/kg)组(UUO+薯蓣皂苷3 d组、UUO+薯蓣皂苷7 d组);薯蓣皂苷组给药3 d或7 d,假手术组和UUO模型组予等体积生理盐水灌胃。HE和MASSON染色观察肾脏病理改变,采用免疫组化法检测肾脏TNF-α的表达。结果:薯蓣皂苷组小鼠肾组织病理损伤程度和TNF-α的表达水平低于模型组。结论:薯蓣皂苷通过抑制TNF-α的表达减轻炎症反应,延缓肾脏纤维化。Objective: To investigate the effects of Dioscin on renal histopathology changes and the expression of TNF-α in unilateral ureteral obstruction(UUO) mice. Methods:Adult male C57BL/6N mice were randomly divided into sham operation group, UUO model group(UUO model 3d, UUO model 7d), Dioscin low-dose(50 mg/kg) group(UUO+Dioscin 3d group, UUO+Dioscin 7d group), high-dose Dioscin(100 mg/kg) group(UUO+Dioscin 3d group, UUO+Dioscin 7d group). The mice in Dioscin group were given Dioscin, the other mice were given physiological saline of equal volumes. The pathology of renal tissue was observed by HE and MASSION staining, and the expression of TNF-α was detected by immunohistochemistry. Results: Diosin alleviated the pathological damage of renal tissue in UUO mice, and the expression of TNF-α was down regulated. Conclusion:Dioscin can alleviate inflammatory response by inhibiting the expression of TNF-α, which delays renal fibrosis in UUO mice.
