摘 要:目的 探讨针刺通过腺苷酸活化蛋白激酶(AMPK)、哺乳动物雷帕霉素靶蛋白(mTOR)、UNC-51样自噬激活激酶(ULK)1/2介导的自噬对脑卒中大鼠神经元的影响。方法 50只SD大鼠随机分为假手术组、模型组、针刺组、3-甲基腺嘌呤(3-MA)组、针刺+5-氨基-4-甲酰胺咪唑核糖核苷酸(AICAR)组,每组10只,除假手术组其他4组采用中动脉栓塞法构建模型,并给予相应治疗。治疗后24 h,对各组大鼠神经功能缺损程度进行评分。结果 与假手术组比较,模型组治疗后神经功能缺损评分、海马组织AMPK、磷酸化ULK1(p-ULK1)/ULK1、微管相关蛋白1轻链3(LC3)B/A、Beclin-1表达升高,mTOR表达降低(P<0.05);与模型组比较,针刺组和3-MA组大鼠神经元损伤、自噬减轻,神经功能缺损评分、海马组织AMPK、p-ULK1/ULK1、LC3B/A、Beclin-1表达降低,mTOR表达升高(P<0.05);针刺+AICAR组治疗后神经功能缺损评分、海马组织AMPK、p-ULK1/ULK1、LC3B/A、Beclin-1表达明显高于针刺组(P<0.05),针刺+AICAR组mTOR表达明显低于针刺组(0.36±0.05 vs 0.79±0.12,P<0.05)。结论 针刺通过抑制AMPK-mTOR-ULK1/2通路介导的自噬,减轻脑卒中大鼠的神经元损伤。Objective To investigate the effect of acupuncture on neurons in stroke rats through autophagy mediated by AMPK,mTOR,and ULK1/2.Methods Fifty SD rats were randomly separated into sham operation group, model group(MCAO),acupuncture(Baihui and Shuigou points, once a day, for 7 d) group, 3-methyladenine(3-MA,autophagy inhibitor, 2 mg/kg) group, acupuncture+AICAR(AMPK activator, 0.5 mg/kg) group(n=10).In 24 h after the last treatment, the severity of neurological deficit was scored.Results The neurological deficit score and expression levels of AMPK,p-ULK1/ULK1,LC3B/A and Beclin-1 in the hippocampus were significantly elevated and the level of mTOR was decreased in the model group than the sham operation group(P<0.05).Compared with the model group, the acupuncture group and 3-MA group had obviously mild neuronal damage, attenuated autophagy, lower neurological deficit score, down-regulated AMPK,p-ULK1/ULK1,LC3B/A and Beclin-1 while up-regulated mTOR expression(P<0.05).The neurological deficit score was notably higher, and the hippocampal levels of AMPK,p-ULK1/ULK1,LC3B/A and Beclin-1 were remarkably higher, but that of mTOR(0.36±0.05 vs 0.79±0.12) was lower in the acupuncture+AICAR group than the acupuncture group(P<0.05).Conclusion Acupuncture can reduce the neuronal damage of stroke rats by inhibiting the autophagy mediated by AMPK-mTOR-ULK1/2 pathway.
