摘  要:目的:观察实验性功能性腹泻(FDr)脾虚证大鼠黏膜上皮离子转运的情况,及白术水提液对其上皮离子转运的影响途径。方法:将Wistar大鼠随机分为正常组和模型组,采用高乳糖饲料喂养叠加小平台站立的复合因素的方法造模,通过体外应用尤斯灌流装置,观察正常组和模型组大鼠的短路电流变化和白术水提液对大鼠远端结肠上皮离子转运所产生的短路电流的作用机制。结果:正常组与模型组的基础电生理特性没有显著性差异;60 mg/mL白术水提液能够引起两组大鼠黏膜上皮短路电流显著下降(P<0.05),分别加入Na^(+)通道阻断剂amiloride、Cl^(-)/HCO3-交换体抑制剂DIDS后,两组大鼠的短路电流没有明显改变;采用非选择性Cl^(-)、K^(+)通道阻断剂,Na^(+)-K^(+)-2Cl^(-)转运体抑制剂DPC、TEA、bumetanide后,两组大鼠的短路电流均显著受到抑制(P<0.05);在无Cl^(-)、无Cl^(-)/HCO3-的KHS中,白术水提液引起的短路电流均减小(P<0.05),但在无HCO3-的KHS液中,短路电流差异无统计学意义。结论:白术水提液对FDr大鼠上皮离子转运的影响是通过调控顶膜侧上的Cl^(-)通道、K^(+)通道及基底膜侧Na^(+)-K^(+)-2Cl^(-)转运体,影响Cl^(-)的吸收、K^(+)的分泌,从而调节结肠上皮的水和电解质的平衡。Objective:To investigate the changes of mucosal epithelial ion transport in rats with functional diarrhea(FDr) spleen deficiency syndrome and the effect of Atractylodes macrocephala aqueous extract on its epithelial ion transport.Methods:The Wistar rats were randomly divided into a normal group and model group.The rat model was made by feeding high lactose feed with compound factors superimposed on small platform.Using Ussing chamber device in vitro,the changes of short-circuit current (Isc) in normal group and model group and the effect of Atractylodes macrocephala aqueous extract on ion transport in distal colonic epithelium of the two groups were observed.Results:There was no significant difference in basic electrophysiological characteristics between normal group and model group.The aqueous extract of Atractylodes macrocephala 60 mg/m L could significantly reduce the short-circuit current of mucosal epithelium in the two groups (P<0.05).After adding Na^(+)channel blocker,Cl^(-)/HCO3-exchanger inhibitor amiloride and DIDS respectively,there was no significant change in Isc in the two groups.After using non selective Cl^(-),K^(+)channel blockers,Na^(+)-K^(+)-2Cl^(-)transporter inhibitors DPC,tea and bumetanide,the Isc of the two groups was significantly inhibited (P<0.05);After replacing Cl^(-),Cl^(-)/HCO3-from KHS solution,the Isc induced by Atractylodes macrocephala aqueous extract decreased (P<0.05),but there was no significant difference in Isc after replacing HCO3-from KHS solution.Conclusion:The effect of Atractylodes macrocephala aqueous extract on epithelial ion transport in and FDr rats may be through regulating Cl^(-)channel,K^(+)channel on parietal membrane and Na^(+)-K^(+)-2Cl^(-)transporter on basement membrane,affecting Cl^(-)absorption,K^(+)secretion,so as to restore the banlance of water and electrolyte transport of colonic epithelium.
