芪黄健脾滋肾颗粒对肾小球系膜细胞过度增殖的改善作用

摘　　要：目的:探讨芪黄健脾滋肾颗粒改善肾小球系膜细胞增殖的作用及其可能机制。方法:以体外培养的HBZY-1肾小球系膜细胞为模型,模型组采用TGF-β1刺激模拟肾小球系膜细胞过度增殖,血清组在模型组基础上加入芪黄健脾滋肾颗粒含药血清;采用CCK-8法检测肾小球系膜细胞的增殖活力;运用RT-PCR技术检测LncRNA GAS5、miR-21-5p、Sprouty1 mRNA、ERK1/2 mRNA的表达;采用Western blot法检测Sprouty1、p-ERK1/2、ERK1/2蛋白表达;结果:含药血清在10%浓度下对模型组的异常细胞增殖有明显的抑制作用(P<0.01);与正常组比较,模型组的ERK1、ERK2 mRNA和磷酸化蛋白及miR-21-5p表达水平均显著升高(P<0.01),GAS5和Sprouty1mRNA的表达则显著降低(P<0.01);与模型组比较,血清组可显著降低ERK1、ERK2 mRNA和磷酸化蛋白及miR-21-5p的表达(P<0.01),以及显著增加GAS5和Sprouty1 mRNA的表达(P<0.01)。结论:芪黄健脾滋肾颗粒可通过上调GAS5的表达,结合mi R-21、增加Sprouty1的表达,降低下游ERK1/2的表达,从而改善肾小球系膜细胞增殖。Objective: To investigate the effects of Qihuang Jianpi Zishen Granules on the proliferation of mesangial cells(MC) and its possible mechanism. Methods: HBZY-1 mesangial cells cultured in vitro were used as the model,in the model group,TGF-β1 was used to stimulate the proliferation of mesangial cells in renal glomerulus,serum group was supplemented with serum containing Qihuang Jianpi Zishen Granules on the basis of model group. CCK-8 assay was used to detect the proliferation of glomerular mesangial cells. The expressions of lncRNA GAS5,miR-21-5p,Sprouty1 mRNA and ERK1/2 mRNA were detected by RT-PCR. The protein expressions of Sprouty1,p-ERK1/2 and ERK1/2 were detected by Western blot. Results: The abnormal cell proliferation in model group was significantly inhibited by serum containing drugs at 10% concentration(P<0.01). Compared with the normal group,the mRNA and protein expression levels of ERK1,ERK2 and miR-21-5p in the model group were significantly increased(P<0.01),while the expression levels of GAS5 and Sprouty1 mRNA were significantly decreased(P<0.01). Compared with model group,the mRNA and protein expression of ERK1,ERK2 and miR-21-5p in serum group were significantly decreased(P<0.01),and the expressions of GAS5 and Sprouty1 mRNA were significantly increased(P<0.01). Conclusion: Qihuang Jianpi Zishen Granules can improve the proliferation of glomerular mesangial cells by upregulating the expression of GAS5,combining with miR-21,increasing the expression of Sprouty1,and reducing the expression of downstream ERK1/2.
