凉血解毒化瘀汤治疗肝衰竭的作用机制

摘　　要：目的:基于网络药理学、分子对接探讨凉血解毒化瘀汤治疗肝衰竭的作用机制。方法:通过中药系统药理学数据库分析平台(TCMSP)查找凉血解毒化瘀汤药物的主要活性成分及其靶点;通过GenCards、TTD数据库获取肝衰竭的相关作用靶点;利用Cytoscape3.8.2软件构建活性成分-疾病靶点的网络图;通过STRING平台进行蛋白质相互作用分析,构建PPI核心网络;通过Bioconductor平台对共同靶点进行功能富集分析及通路分析。结果:经过数据库筛选共获得凉血解毒化瘀汤治疗肝衰竭的活性成分为槲皮素、β-谷甾醇、木犀草素等,核心靶点为TP53、TNF、STAT3等。通过富集分析可知主要通过脱氧核糖核酸-结合转录、核受体活性配体-激活、转录因子活性蛋白酶结合等方式参与对异生素、脂多糖、细菌源性分子及氧水平的反应等生物进程。结论:凉血解毒化瘀汤可通过多成分-多靶点-多通路治疗肝衰竭。Objective: To explore the mechanism of Liangxue Jiedu Huayu decoction in treating liver failure based on network pharmacology and molecular docking. Methods: The main active ingredients and targets of Liangxue Jiedu Huayu Decoction were searched through the database analysis platform of traditional Chinese medicine system pharmacology(TCMSP);the relevant targets of liver failure were obtained through GenCards and TTD databases;Cytoscape3.8.2 software was used to construct Active ingredient-disease target network diagram;protein interaction analysis through STRING platform to construct PPI core network;functional enrichment analysis and pathway analysis of common targets through Bioconductor platform. Results: After database screening, the active ingredients of Liangxue Jiedu Huayu Decoction in the treatment of liver failure were quercetin, β-sitosterol, luteolin, etc., and the core targets were TP53, TNF, STAT3, etc. Through enrichment analysis, it can be known that it mainly participates in the response of xenobiotics, lipopolysaccharides, bacterial-derived molecules and oxygen levels through DNA-binding transcription, nuclear receptor active ligand-activation, transcription factor activity, protease binding, etc. process. Conclusion: Liangxue Jiedu Huayu Decoction can treat liver failure through multiple components-multiple targets-multiple pathways.
