四味黄连洗剂对白色念珠菌和近平滑假丝酵母菌抑菌作用研究及有效成分筛选

摘　　要：目的:讨论四味黄连洗剂提取物体外对真菌的抑菌作用;明确四味黄连洗剂的主要抑菌药材和成分。方法:以K-B法药敏实验体外测定四味黄连洗剂对白色念珠菌、近平滑假丝酵母菌代谢活性的影响,探讨四味黄连洗剂有效药味并进行缺味验证实验;CLSI法测定四味黄连洗剂的最低MIC值;CLSI和Spot assay法测定盐酸小檗碱的最低MIC值及对近平滑假丝酵母菌细胞活性的影响。结果:四味黄连洗剂对近平滑假丝酵母菌有明显抑制;对近平滑假丝酵母菌最低抑菌浓度为128μg/mL;黄连、黄柏为有效抑菌药材;有效成分盐酸小檗碱对近平滑假丝酵母菌的MIC值为64μg/mL,对近平滑假丝酵母菌细胞活性有较大影响。结论:该研究表明,四味黄连洗剂对近平滑假丝酵母菌有抑制作用,且其主要有效抑菌成分为盐酸小檗碱。Objective:To discuss the antibacterial effect of Siwei Huanglian lotion extract on fungi in vitro;to clarify the main antibacterial medicinal materials and components of Siwei Huanglian lotion.Methods:The effect of Siwei Huanglian lotion on the metabolic activity of Candida albicans and Candida parapsilosis was determined in vitro by K-B method drug susceptibility test,and the effective taste of Siwei Huanglian lotion was explored and the lack of taste verification experiment was carried out;CLSI method was used to determine the four The minimum MIC value of Siwei Huanglian lotion;CLSI and Spot assay method to determine the minimum MIC value of berberine hydrochloride and its effect on the cell viability of Candida parapsilosis.Results:Siwei Coptidis lotion had significant inhibitory effects on Candida parapsilosis;the minimum inhibitory concentration on Candida parapsilosis was 128μg/mL;Coptis chinensis and Phellodendron Phellodendri were effective antibacterial medicinal materials;the active ingredient berberine hydrochloride was effective against The MIC value of Candida parapsilosis is 64μg/mL,which has a great influence on the cell viability of Candida parapsilosis.Conclusion:This study shows that Siwei Huanglian lotion has inhibitory effect on Candida parapsilosis,and its main effective antibacterial component is berberine hydrochloride.
