摘  要:目的探究纳米雄黄引起B细胞非霍奇金淋巴瘤Raji细胞凋亡和自噬的作用机制。方法将细胞设为对照组和给药组(给药组分为水飞雄黄组和纳米雄黄组),进行CCK-8细胞活力检测、荧光显微镜观察、流式细胞术检测以及Western blot实验。结果与对照组相比,水飞雄黄组和纳米雄黄组均会抑制Raji细胞的增殖,且呈浓度依赖性,其中,纳米雄黄组的抑制作用更为显著。Western blot结果表明,纳米雄黄药物处理Raji细胞24 h后,凋亡相关蛋白Bax表达升高,Bcl-2蛋白表达下降;自噬相关蛋白Beclin-1表达升高,p62蛋白表达下降。结论纳米雄黄可以通过线粒体途径诱导Raji细胞发生凋亡,且能够诱导Raji细胞发生自噬,有效抑制细胞增殖从而发挥抗肿瘤作用。Objective To determine the mechanism of nano-realgar on the apoptosis and autophagy of B cell non-Hodgkin’s lymphoma Raji cells.Methods The cells were divided into a control group,a crude realgar group and a nano-realgar group.The CCK-8 cell viability assay,fluorescence microscopy,flow cytometry and Western blot were performed.Results Compared with the control group,the proliferation of Raji cells was inhibited in a concentration-dependent manner in both the crude realgar group and the nano-realgar group.The inhibition effect in the nano-realgar group was more obvious.Western blot showed that Bax expression was increased and expression of Bcl-2 protein decreased after the treatment with Raji cells for 24 h.The expression of autophagy-related protein Beclin-1 was elevated,while p62 protein expression decreased.Conclusion Nano-realgar can inhibit the proliferation of Raji cells,and induce their apoptosis and autophagy,which may be related to the inhibition of the expression of related pathway proteins.
