摘 要:为了探讨不同浓度的羧甲基壳聚糖(carboxymethyl chitin,CMCS)对猪精液常温保存效果的影响,试验采集了4头健康的2~3岁长白猪精液,利用未添加CMCS的Modena稀释液和添加不同浓度(30,50,70,100μg/mL)CMCS的Modena稀释液进行稀释,然后将稀释的猪精液在17℃环境中保存5 d,于保存第0(添加当天),1,3,5天测定精液精子活力、线粒体活性率、质膜完整率、精液pH值和渗透压指标,筛选CMCS的最佳添加浓度。结果表明:在常温保存过程中添加CMCS对猪精液具有保护作用,在精液保存第5天时,添加70μg/mL CMCS精液的精子活力、线粒体活性、质膜完整率最高,显著高于未添加和其他浓度(P<0.05);添加70μg/mL CMCS精液的渗透压最高,显著高于100μg/mL浓度(P<0.05),与未添加和30,50μg/mL浓度差异不显著(P>0.05);添加30,50,70μg/mL CMCS精液的pH值差异不显著(P>0.05),但均显著高于未添加和100μg/mL浓度(P<0.05)。说明CMCS在猪精液常温保存中的适宜添加浓度为70μg/mL。In order to investigate the effects of carboxymethyl chitin(CMCS)at different concentrations on the preservation of pig semen at room temperature,semen was collected from 4 healthy 2-3-year-old landrace pigs.The semen was diluted with Modena diluent without CMCS and Modena diluent with different concentrations(30,50,70,and 100μg/mL)of CMCS.Then the diluted pig semen was stored at 17℃for 5 days.Sperm motility,mitochondrial activity rate,plasma membrane integrity rate,semen pH value and osmotic pressure were measured on day 0(add the day),1,3,5 after semen storage,and the optimal concentration of CMCS was screened.The results showed that the addition of CMCS had protective effects on pig semen during the storage of pig semen at room temperature.On the 5th day of semen storage,the sperm motility,mitochondrial activity and plasma membrane integrity rates in the semen supplemented with 70μg/mL CMCS were the highest,which were significantly higher than those in other groups(P<0.05).At this point,the osmotic pressure of semen was also the highest,which was significantly higher than that in the concentration 100μg/mL group(P<0.05),and the difference was not significant compared with that in the group without CMCS,30μg/mL group and 50μg/mL group(P>0.05).There was no significant difference in pH value of semen among the groups supplemented with 30,50 and 70μg/mL CMCS(P>0.05),but pH value in the groups was significantly higher than that in the non-added group and 100μg/mL CMCS group(P<0.05).The results indicated that the optimal concentration of CMCS in pig semen preservation at room temperature was 70μg/mL.
